母体血浆中DNA片段中鸟嘌呤-胞嘧啶(GC)含量相关的PCR扩增错误是产前筛查三聚体错误的原因之一。本研究描述了一个简单的方法可以避免此类错误的发生。

研究人员通过对Reflex数据筛选项目(67三倍体1883影响怀孕)来比较18号染色体DNA片段的比例数8号染色体DNA片段数量(因为染色体8也有类似的GC含量染色体18)18号染色体DNA计数的百分比使用分母中所有常染色体的计数,并对DNA片段的GC含量进行所有常染色体校正。

研究显示,18号染色体与8号染色体的DNA片段计数比率比18号染色体产生的所有常染色体计数的百分比具有更大的识别性,而18号染色体没有或存在GC含量偏差的所有常染色体校正。它达到了很高的筛选性能,如对于0.25%的假阳性率,不进行GC含量校正的检出率为97%,而不进行校正则为49%,对GC含量进行全常染色体校正的检出率为91%

研究表明,在细胞游离DNA产前筛查18号染色体三体时,可以考虑使用18号染色体DNA片段计数与8号染色体DNA片段计数的比值,避免对目前使用的GC含量进行更复杂的校正方法。
