树突状细胞在系统性红斑狼疮中的作用机制新发现】一项新的研究为自身免疫性疾病,即系统性红斑狼疮的潜在发病机制提供了有趣的见解。

该研究发表于12月的Cell Press上的Immunity杂志上,研究展示了在系统性红斑狼疮(systemic lupus erythematosus, SLE)和其他自身免疫性疾病中起关键作用的一种免疫细胞——树突状细胞(Dendritic cells, DCs),以期将树突状细胞作为一种新的治疗靶点。

系统性红斑狼疮是一种慢性自身免疫性疾病,该病可以累及全身多个器官和组织,从而导致各种不同的临床表现。同其他自身免疫性疾病一样,在系统性红斑狼疮中,免疫系统攻击的对象是机体自身的细胞和组织。之前的研究显示,系统性红斑狼疮疾病的发生和获得性免疫中的两种细胞的活化有关,即B细胞和T细胞。

来自康涅狄格州,纽黑文,耶鲁医学院的高级研究作者Mark J. Shlomchik博士说:“我们对机体中另一种类型的免疫细胞产生兴趣,即树突状细胞,并研究树突状细胞在系统性红斑狼疮发病中所起的作用。传统认为,树突状细胞主要触发和控制针对感染的获得性免疫应答,那么树突状细胞具有潜力通过不同的方式影响系统性红斑狼疮的发病。”

Shlomchik博士和他的同事检测了系统性红斑狼疮老鼠模型中的树突状细胞,结果发现,树突状细胞有助于T细胞的扩增和分化,但是令人惊奇的是,T细胞的初始活化并不需要树突状细胞的参与。

这些发现是令人意想不到的,因为已经建立的学说认为,树突状细胞能够触发T细胞和B细胞针对抗原的获得性免疫应答。恰恰相反的是,该研究发现树突状细胞在炎症细胞,如T细胞对靶器官的侵袭过程中起非常重要的作用。树突状细胞缺陷的系统性红斑狼疮鼠模型的肾脏损害和皮肤病变显着减少。树突状细胞还能够显着影响系统性红斑狼疮中产生的自身抗体的数量和质量。

总而言之,该研究表明树突状细胞在自身免疫性疾病中的作用机制和其在针对抗原的一般炎症获得性免疫应答中的作用完全不同。

Shlomchik.博士总结说:“我们的研究显示,在系统性红斑狼疮动物模型中,树突状细胞的作用不是触发而是放大疾病的进程,而该作用和树突状细胞在感染后的免疫应答过程中的作用机制并不相同。虽然在确定树突状细胞在自身免疫性疾病中的具体作用机制上,我们未来还会有很多的工作需要做,但是我们目前的数据已经证实树突状细胞可以作为自身免疫中的一个新的治疗靶点,未来我们将继续研究树突状细胞如何促进局部炎症的进行,以及检测清除树突状细胞是否可以用来治疗疾病。”
