明尼苏达大学发现药物成瘾治疗的新靶标--星形胶质细胞

明尼苏达州大学医学院的一项研究发现了一种药物成瘾的新型潜在靶标。多巴胺是大脑的主要奖励分子之一,有助于学习、记忆和激励行为。多巴胺的破坏与成瘾相关的疾病有关,例如苯丙胺类物质的使用和滥用。《Neuron》上发表的一项最新研究表明,靶向星形胶质细胞钙信号传导可能会降低苯丙胺的影响。该研究由明尼苏达大学医学院Michelle Corkrum博士和Ana Covelo博士共同完成。

传统意义上,星形胶质细胞被认为是大脑的支持细胞,但这项研究表明,星形胶质细胞确实有助于信息处理。Corkrum及其同事发现,星形胶质细胞对多巴胺有反应,其钙的含量增加。钙的增加与ATP /腺苷的释放有关,以调节神经活动。然后,他们专门研究了苯丙胺,因为已知苯丙胺会增加生物体内的多巴胺含量和精神运动活性。他们发现星形胶质细胞对苯丙胺的反应随着钙的增加而增加,如果星形胶质细胞的活性被消除,小鼠模型中苯丙胺的行为作用就会降低。
