当寨卡病毒横空出世时科学家们对其知之甚少,在过去一年里研究者们通过深入研究来阐明寨卡病毒为何会引发一系列危险的健康问题,包括新生儿缺陷等,比如小头畸型和神经性问题(格-巴二氏综合征等),但研究者们并不知道寨卡病毒哪种蛋白会让其变得具有杀伤性,当然他们也不清楚这些蛋白质的作用机理是什么。

近日,一项刊登在国际杂志PNAS上的研究报告中,来自马里兰大学医学院的研究人员通过研究首次在寨卡病毒中鉴别出了让病毒变得具有杀伤性的关键蛋白质,文章中研究人员也首次对寨卡病毒的基因组进行了深入分析。研究者Richard Zhao教授说道,该病毒的作用机制一直是个谜,本文研究为我们深入阐明寨卡病毒影响细胞的分子机制提供了新的线索,当然对于后期开发新型疗法也至关重要。

在全球范围内寨卡病毒感染了成千上万人,其中很多患者都是美国人,在美国及其属地范围内,截至目前有超过3.8万寨卡病毒感染患者,其中大部分都在波多黎各;而且目前并没有针对寨卡病毒的疫苗或预防性措施。为了检测寨卡病毒,研究者Zhao及其同事利用裂殖酵母进行研究,裂殖酵母能够帮助检测病原体如何影响细胞,其最早用来制造啤酒,经过数十年来,裂殖酵母如今已经被很多科学家用来研究细胞的行为机制了。

研究者Zhao是利用裂殖酵母模型来研究HIV以及大麦黄矮病毒的先驱科学家,因此他对裂殖酵母非常熟悉;在这项研究中,研究者对寨卡病毒的14种蛋白和小型肽类进行分离,随后将酵母细胞暴露于每一种蛋白中,来观察细胞的变化,研究者发现,其中有7种蛋白能够以特殊的方式损伤酵母细胞,抑制细胞的生长并且杀灭酵母。

当然研究者的研究并不止步于此,他们还希望后期继续对寨卡病毒进行大量工作,他们希望通过研究理解上述新发现的7种蛋白质在人类机体中的作用机制,目前研究者正在进行初期研究来阐明寨卡病毒同大鼠和人类细胞的相互作用,后期他们希望通过联合研究阐明寨卡病毒感染机体的更多分子机理。
