研究者Matthew Thomson说道,我们发现了一种基本的机制,细胞可以利用该机制来决定是否进行发育;在胚胎发育期间,干细胞会表演一段精心安排的“舞蹈”,随后其会从无作用、未分化的形式转化成为构建机体主要器官系统的细胞。近些年来科学家们在未分化的干细胞中发现了很多可以编码干细胞发育的基因,而揭示这些细胞如何忽视嘈杂的波动以及快速反应形成机体所需细胞一直是科学家们的研究热点。

为了检测干细胞如何将发育线索作为关键的信号或是外部“噪音”,科学家们对培养中的小鼠胚胎干细胞进行了工程化操作,他们利用蓝色光脉冲开启了一种名为Brn2的基因,该基因是一种潜在的神经分化的线索,通过调整光脉冲的强度和持续性,研究者就可以实现精确控制Brn2的剂量,并且观察细胞的反应。研究者表示,如果Brn2信号足够强的话,干细胞就会快速转化成为神经元。

随后研究者利用CRISPR-Cas9基因编辑技术给一种名为Nanog的转录因子添加上了一个荧光探针,这样在正常情况下其就可以扮演干细胞分化的制动器了,Nanog就可以被用来读取细胞决策制定的情况。转录因子Nanog本身就是一种时间调节器,当Brn2信号开启时,其就会干扰分子回路来保持细胞处于稳定和未分化的状态,而Nanog的水平就会随之降低;如果如果Nanog用尽而Brn2信号持续存在的话,就会使得干细胞快速转化成为神经元。

最后研究者Thomson说道,细胞并不是傀儡,其是一种可以持续解释信息的的介值,就好比大脑一样;如果我们想精确地操控细胞命运,那么我们必须理解细胞中信息处理的机制,并且理解这种机制如何发生。
