bioRxiv:科学家成功分离高效抗新冠病毒抗体

3 月 27 日,清华大学宣布,该校与深圳市第三人民医院合作,成功分离了高效抗新冠病毒抗体,为治疗和预防新冠肺炎提供强有力的医学干预手段。该项成果已投《自然》并发表在预印本平台 bioRxiv。

自新冠肺炎疫情暴发以来,清华大学医学院教授张林琦团队与深圳市第三人民医院教授张政团队合作,对新冠病毒感染和康复期病人体内保护性抗体反应进行了全面分析。

该团队利用抗体分析技术、抗体分离技术和抗体评估技术,从新冠肺炎康复者血液的 B 淋巴细胞中,分离出 200 多株抗新冠病毒的单克隆抗体及其编码基因。这些抗体是康复者在病毒感染过程中产生的天然武器,可以有效识别出病毒表面蛋白的关键靶位,阻止病毒进入人体细胞。

据介绍,在人体血液中,抗体可以占到全部血浆蛋白的 20%,并随着血液的流动在全身持续不断地巡视外来入侵病毒,并对其实施强大的抑制和清除作用。这也是人体能够产生免疫力的物质基础。人体抗体由 B 淋巴细胞产生,且每个抗体都有自己的特点。

“如果我们把抗体比喻成一支部队的话,‘多克隆抗体’相当于混合兵种,其能力以及对应的病毒位置有很大差别。有的只是袭击病毒的‘胳膊’,有的则可以识别并攻击病毒的‘心脏’。”张政说,他们的工作便是找到能精准打击新冠病毒 “心脏” 的单个 “士兵”,即所谓的“单克隆抗体”,并获得其基因,进而实现对该“士兵” 的大规模复制生产。

目前,该团队正在与清华大学生命科学学院教授王新泉合作,针对相关抗体开展系统的结构和功能研究,筛选所获抗体中最优秀个体,全力推动大规模生产、临床前动物研究和后续的临床应用开发,为治疗和预防新冠肺炎提供医学干预手段。
