JCI:华人女科学家揭示影响肝癌发生的重要信号网络

近日,来自美国哈佛牙医学院的华人科学家Yingzi Yang带领研究团队对影响肝脏肿瘤发生的信号途径网络进行了深入研究,他们发现包括Hippo, Wnt/b-catenin和Notch等多条信号途径在内的信号网络能够通过相互作用影响肝癌的发育。该研究为肝癌治疗方法的开发提供了重要信息。恶性肿瘤的发育需要经过起始和进展等多个步骤,了解肿瘤起始过程对于肿瘤预防、诊断和治疗都有非常重要的意义。但


近日,来自美国哈佛牙医学院的华人科学家Yingzi Yang带领研究团队对影响肝脏肿瘤发生的信号途径网络进行了深入研究,他们发现包括Hippo, Wnt/b-catenin和Notch等多条信号途径在内的信号网络能够通过相互作用影响肝癌的发育。该研究为肝癌治疗方法的开发提供了重要信息。

恶性肿瘤的发育需要经过起始和进展等多个步骤,了解肿瘤起始过程对于肿瘤预防、诊断和治疗都有非常重要的意义。但是限制正常细胞增殖以及防止肿瘤起始的信号网络还没有得到充分了解。

在这项研究中,研究人员报告称Hippo, Wnt/b-catenin和Notch这几条信号途径形成了一个相互作用的网络来共同维持肝脏尺寸并抑制肝细胞癌的发生。当肝脏细胞缺少Hippo信号途径中的激酶Mst1和Mst2就会导致肝细胞癌的快速形成并激活YAP/TAZ,STAT3,Wnt/b-catenin和Notch信号途径。

之前工作已经发现这些下游通路的异常激活会导致肝细胞癌的发生。进一步的实验表明Notch信号途径会与Hippo信号途径的效应因子YAP/TAZ形成一个正反馈回路促进严重的肝肿大以及肝细胞癌的快速起始和进展。非常令人意外的是,他们还发现Wnt/b-catenin信号途径的激活能够通过抑制YAP/TAZ和Notch信号途径形成的正反馈回路抑制肝细胞癌的形成。

研究人员还发现当肝细胞中缺少了Mst1和Mst2,STAT3对于肝细胞癌的形成变得可有可无。该研究发现的分子网络为研究肝细胞癌的分子归类以及开发治疗策略用以治疗多个基因突变引起的肝脏肿瘤提供了重要见解。
