Science Advance:3D打印再创佳绩!连心脏也能打!

“我们已经能够利用冠状动脉的MRI图像以及胚胎心脏的三维图像,通过3D生物打印,以一种空前的分辨率和质量制造出很软的材料如胶原蛋白、海藻酸盐和纤维蛋白。”卡内基梅隆大学的生物医学工程系材料科学和工程学副教授Adam Feinberg说。Feinberg所带领的再生生物材料和治疗团队的最新研究成果发表在10月23日出版的Science Advances杂志上。

“由于Feinberg教授在生物打印研究上的杰出工作,我们CMU的研究人员继续开发新的方案来解决这样的问题,这将对社会产生变革性的影响,”卡内基梅隆大学工程学院的院长Jim Garrett说,“我们应该期望看到3D生物打印继续发展,并将成为能够大量应用的一个重要的医学工具。”

传统的3D打印机通常利用塑料或金属来制造坚硬的东西,而他们的工作原理是利用把原料一层一层沉淀到物质表面的技术来创建3D物质。每一层的打印均需要坚固的下层作为支持,因此打印软凝胶等材料的能力是有限的。

“在过去的十年中,3D打印各种材料一直是组织工程中的一个常见的发展趋势,但直到现在,还没有人发明了一种装配方法来组合工程凝胶如胶原蛋白或纤维蛋白,”该研究的第一作者、卡内基梅隆大学生物医学工程研究生J Hinton说。

“软材料的挑战类似于对我们所吃的果冻的思考,当在空气中对其进行3D打印时将出现他们在自身重压之下发生坍塌,”Feinberg解释道,“所以我们开发了一个方法使得这些软材料在‘支持’物质中进行打印。从本质上讲,我们在一个凝胶内打印另一个凝胶,因此我们能够准确把握软材料印制的位置,一层一层进行打印。”

这项技术的重大进展之一,称为FRESH,即“自由的悬浮水凝胶的可逆的嵌入”,能够支持易于融化的凝胶,可以被加热到体温,但又不破坏脆弱的生物分子或生物打印出来的活细胞。下一步,该研究团队将致力于将真正的心脏细胞纳入这些3D打印组织结构,利用这个支架的帮助形成具有收缩功能的肌肉。

生物打印是一个先进的领域,但迄今为止,大多数3D生物打印花费超过100000美元,并且操作需要专业知识,这限制了这种技术的广泛应用。而今,Feinberg的研究团队已经能够在客户级3D打印机上实现他们一系列的技术,花费不到1000美元并且利用的是开放获取的硬件和软件。

“不仅是成本低,而且通过利用开放获取的软件,我们能够自主调整印刷参数,优化我们的成品,使得我们打印的质量达到最大化”Feinberg说,“这真切促使我们加快发展新材料和拓展创新的空间。同时,我们也将通过发布我们的3D打印设计理念来回馈开放获取的资源。”
