背景和目的

方法
研究人员使用Illumina阵列在11个UC-CRC和邻近组织以及11个散发性CRC和邻近组织中评估DNA甲基化程度。然后对10个UC-CRC和邻近组织以及8个散发性CRC和邻近组织进行RNA测序,评估了DNA甲基化和转录物表达的差异,以及它们在相同组织中的相关性。最后研究人员对三种蛋白质ANPEP,FAM92A1和STK31进行免疫组织化学分析。

结果
本项研究显示33个基因座显示出UC-CRC和邻近组织之间DNA甲基化的差异。相反,散发性结肠癌和邻近组织之间存在4204个差异甲基化位点。886个基因以及10个长的非编码RNA [lncRNA]在UC-CRC和相邻组织之间差异表达。虽然UC和散发性CRC之间没有差异甲基化位点,但UC-CRC和散发性CRC之间有997个基因和38个lncRNA差异表达。在UC中,18个基因表现出DNA甲基化和转录物表达之间的负相关。

结论
本项研究表明机体通过DNA甲基化调节转录物表达导致相关不同结肠癌的发生。炎性肠病和散发性结肠癌之间的表现和结果是存在差异的。
