ANN RHEUM DIS:信号素4D抑制中性粒细胞活化并参与中性粒细胞介导的自身免疫性血管炎

中性粒细胞表面SEMA4D作为中性粒细胞活化的负性调节因子。

中性粒细胞的异常活化在抗中性细胞胞浆抗体(ANCA)相关性血管炎(AAV)中起重要作用。近日风湿病权威杂志ANN RHEUM DIS上发表了一篇文章,旨在阐述信号素 4D(SEMA4D)在调节中性粒细胞活化及其参与AAV发病机制中的功能。

采用ELISA法检测可溶性SEMA4D的血清水平。通过流式细胞仪检测血细胞表面的膜SEMA4D的表达。为明确中性粒细胞膜SEMA4D和内皮丛细胞B2之间的相互作用,野生型和SEMA4D - / -小鼠中性粒细胞与用SYTOX绿染色的内皮细胞系(MS1)一起培养,并进行中性粒细胞胞外捕获(NET)形成测定。通过测量动力学氧化爆发和NET形成测定来评估用重组plexin B2处理人中性粒细胞的功效。

AAV患者血清SEMA4D水平升高,与疾病活动评分相关。SEMA4D的细胞表面表达在AAV的中性粒细胞中下调,这是膜SEMA4D的蛋白水解切割的结果。可溶性SEMA4D对内皮细胞产生促炎作用。中性粒细胞上的膜SEMA4D与内皮细胞上的丛状蛋白B2结合,这种相互作用降低了NET形成。重组plexin B2通过SEMA4D的细胞内结构域抑制中性粒细胞Rac1的活化,并抑制病原体诱导或ANCA诱导的氧化爆发和NET形成。

由此可见,中性粒细胞表面SEMA4D作为中性粒细胞活化的负性调节因子。在AAV患者中观察到的SEMA4D的蛋白水解切割可能会促进中性粒细胞介导的炎症反应。SEMA4D可作为AAV的生物标记物和潜在的治疗靶标。
