Nat Med:一种小分子可增强膜蛋白活性或可治疗疼痛

KCC2相当于一台跨膜泵,可让神经细胞中的氯离子浓度保持在低水平,从而抑制神经冲动。因此,Yves De Koninck等人推断,能够激活KCC2的药物或许也能从临床水平上修复抑制信号并治疗神经疾病。他们筛选了一类小分子化合物库并确定一种他们成为CLP257的药物。然后,研究人员进一步发现这种药物能够增强KCC2的表达和活性,并减少神经损伤大鼠的疼痛。

他们注意到另一种相关化合物在药理学方面更胜过CLP257,该化合物在减轻大鼠疼痛方面与现有的一种治疗疼痛的药物pregabalin具有同样的效果,却不会像pregabalin那样对运动神经产生多余的副作用。
