中国科学家全球首建自闭症猴模型

如何对自闭症患者进行药物治疗,至今仍是个难题。要解开这道难题,第一步是有可供研究的动物模型和试验平台。

1月26日,英国《自然》杂志在线发表了中科院上海生科院神经科学研究所(以下简称神经所)仇子龙研究组与该所苏州非人灵长类研究平台孙强团队的合作成果。研究中,他们通过遗传学手段让猴子患上了自闭症,这一成果标志着中国科学家在世界上首次建立了携带人类自闭症基因的非人灵长类动物模型。

为啥要让猴子“自闭”?

自闭症,又被称为孤独症,是青少年多发的一类发育神经性精神疾病。多年来,科学家对自闭症的研究早已从临床病理层面深入到细胞以及分子的病理层面。

在这一过程中,科学家发现了一个名叫MeCP2的基因。该基因被认为是导致自闭症的“罪魁祸首”。MeCP2基因可间接对神经功能造成影响。MeCP2在体内的表达量就像一座天平,必须保持精妙的平衡,表达过多或过少,都会导致神经突触及神经系统功能异常,从而导致自闭症。

已有研究显示,MeCP2基因功能的缺失会导致瑞特综合征,该病症发于女童,患者表现出类自闭症的行为特征;而当含有MeCP2基因的染色体区段发生拷贝数倍增时,则会导致MeCP2倍增综合征,让男性患者表现出严重自闭症症状。

科学家将目光集中在MeCP2上,并构建了MeCP2过表达的小鼠,作为研究自闭症的动物模型。但仅有转基因小鼠模型还不够,“自闭症涉及多种复杂的高级神经活动,如果要为临床研究与治疗提供更有力支持,小鼠研究已捉襟见肘,更高级的动物模型和更复杂的神经系统非常必要。”仇子龙说。

猴子是怎么“自闭”的?

为了得到更高级的动物模型和更复杂的神经系统,仇子龙研究组和孙强团队决定尝试让非人灵长类动物——食蟹猴,携带人类的MeCP2基因。

他们首先通过使用慢病毒载体侵染方法,将外源基因带入猴子体内,建立了在神经系统中特异性过表达MeCP2基因的转基因食蟹猴模型。然后通过序列捕捉、深度测序及分析定位,发现外源的人类MeCP2基因有效地插入了食蟹猴的基因组中,且免疫印迹及免疫组织化学染色证据显示,人类MeCP2转基因能够在食蟹猴神经系统中产生特异性表达。

自闭症的“种子”种下后,会在食蟹猴身上产生怎样的作用呢?带着这样的疑问,研究人员对此模型进行了长期的体征观察和大量的行为学测试。

通过与野生型对照组对比,他们发现,转基因猴体重发育迟缓,脂肪酸代谢出现异常;在行动路线的追踪中,转基因组相较于对照组会明显花费更多时间在重复的行动路线上;在焦虑应激的实验中,转基因组比对照组也表现出明显的焦虑与警惕。

最重要的是,在与社交相关的行为学实验中(包括社群中和配对的社交时间分析中),转基因猴的社交时间均显着低于对照组。而在与学习记忆相关的测试中,转基因组虽然没有显示出学习能力异常,但表现出重复的刻板行为。这些结果与MeCP2倍增综合征患者的临床表型非常相似。

与此同时,孙强等人通过精巢异体移植的方法,成功地得到了携带人类MeCP2基因的第二代转基因猴。“这说明自闭症作为一种遗传性疾病,在转基因猴模型中得到了很好的体现。”孙强说。

得了自闭症之后呢?

让猴子患上自闭症当然不是科学家们的最终目的。仇子龙告诉记者,目前研究人员已经在对患有自闭症的转基因猴进行详细的脑成像研究,以期待用基因编辑工具对这些携带自闭症基因的转基因猴进行基因治疗,探索最终治愈自闭症之路。

国际权威评审专家认为,此项研究描述了用慢病毒转基因方法,得到了携带人类自闭症相关基因MeCP2的转基因食蟹猴,并对这些动物的行为与认知能力进行了详尽的研究,发现MeCP2转基因猴表现出类似人类自闭症患者的行为特征。

“研究具有原创性与重要性,首次建立了携带人类自闭症基因的非人灵长类动物模型,为深入研究自闭症的病理与探索可能的治疗干预方法提供了重要基础。”一位评审如是评价。

中科院脑科学与智能技术卓越创新中心主任、神经所所长蒲慕明也表示,这项工作为观察自闭症的神经科学机理研究提供了一扇重要窗口,转基因食蟹猴作为自闭症的非人灵长类模型为更深入研究自闭症病理及探索治疗干预方法奠定了坚实基础,最终为临床治疗提供了良好的动物模型和试验平台,表明中国已成为全世界非人灵长类疾病动物模型的研究中心。
