Nature: 科学家为姜黄素“正名”

姜黄素是从姜科、天南星科中的一些植物的根茎中提取的一种化学成分。医学研究表明,姜黄素具有降血脂、抗肿瘤、抗炎、利胆、抗氧化等作用。姜黄素的临床应用潜力,不应该简单地因为其在分子药物筛选中产生的困惑结果而被摒弃

姜黄素是从姜科、天南星科中的一些植物的根茎中提取的一种化学成分。医学研究表明,姜黄素具有降血脂、抗肿瘤、抗炎、利胆、抗氧化等作用。姜黄素的临床应用潜力,不应该简单地因为其在分子药物筛选中产生的困惑结果而被摒弃(Nature,541,144–145;2017K. M. Nelson,et al.J Med Chem. http://doi.org/bw46;2017)。

Nelson及其同事宣称,尽管有上千的研究文章和超过120个临床试验(www.clinicaltrials.gov),姜黄素的治疗益处仍然缺乏证据。然而,通过PubMed搜索“curcumin double-blind placebo-controlled clinical trial(姜黄素,双盲安慰剂对照临床试验)”,有49个结果,其中17个最近的试验显示姜黄素是有效的;另外,还有27个临床试验和至少5个动物实验也指出了姜黄素的益处。

候选药物必须有单一的已知靶点,并且与高通量筛选具有兼容性的假设,阻止了有价值的候选药物进入临床(RL Elliott, Am Chem Soc Med Chem Lett. 3,688-690; 2012)。目前检测靶点的方法不能测量研究药物的完整药理学谱,因此应该与其它筛选方法一起使用。而且,姜黄素对多个分子靶点的结合行为与调节作用,而不是直接抑制相关。高通量筛选容易产生技术上的假象,使其成为排除潜在药物的欺骗性独断者。

考虑到上述情况,如果我们要建立能在人类和动物上应用的有价值的结果,姜黄素分子靶点和调控机制仍需要更深入的研究。
