D2d的设计来自国家糖尿病、消化系统病和肾病研究所(NIDDK,National Institute of Diabetes and Digestive and Kidney Diseases) U34项目资助。最终协议由D2D研究组、数据安全监察委员会(the data and safety monitoring board)和NIDDK批准。


关键的入组标准为年龄大于等于30岁,体重指数为24–42 kg/m2(亚裔美国人为22.5),糖尿病的风险增加[定义为满足美国糖尿病协会制定的糖尿病前期的3个血糖标准中的2个:空腹血糖(FPG) 为100–125 mg/dL(5.5–6.9 mmol/L),75g葡萄糖负荷后餐后2小时血糖值140–199mg/dL(7.7–11 mmol/L),糖化血红蛋白值5.7–6.4%(39–46 mmol/mol)],无甲状旁腺功能亢进症,肾结石或高钙血症。

D2d参与者被随机分为每日一次的维生素D3(胆骨化醇4000 IU)或安慰剂治疗,平均随访3年。主要终点是发生糖尿病的时间,采用研究中的标准实验室评估或D2d以外的诊断。招募在2013年底启动。
