在发育中的大脑内,新的神经元是由神经源性(neurogenic)区域中的神经干细胞产生的,在这个区域中,这些干细胞被指示何时和多久它们应当分裂或它们应当产生什么类型的后代细胞。然而,在此之前,负责提供这些指示的信号一直是不清楚的。

论文第一作者、伦敦大学学院眼科学研究所科学家Mathew Tata解释道,“我们发现血管在告诉神经干细胞何时和如何增殖中发挥着一种关键性的作用。我们研究缺乏血管蛋白NRP1的小鼠脑干中的神经干细胞行为,其中大脑中的这一部分在控制呼吸和心率等基础过程中发挥着特别重

要的作用。在脑干的神经源性区域阻止血管生长会干扰正常的神经元产生,导致神经干细胞丢失它们的增殖能力。因此,在脑干的生长完成之前,神经干细胞从脑干中消失,因此缺乏NRP1的小鼠最终拥有更小的脑干。”

这项研究提供首个证据证实血管不仅对运送血液到发育中的大脑内是比较重要的,而且也在干细胞信号转导中发挥着重要作用。

论文通信作者、伦敦大学学院眼科学研究所科学家Christiana Ruhrberg说,“血管因它们在供应氧气和营养物给大脑中所发挥的重要功能而被人广为所知。然而,这项研究最为吸引人的发现是血管并不是简单地通过它们在大脑氧合作用中的作用或者让大脑组织保持健康来调节脑干中的神经干细胞行为。我们发现血管也提供重要的信号来让神经干细胞在永久性地变成不能增殖的神经细胞之前在较长的时间内增殖。”
