这项研究发表在11月7日的《细胞》(Cell)杂志上。
论文的资深作者、神经生物学助理教授Valeria Cavalli博士说:“我们知道有几个基因可促成称之为轴突的这些神经细胞分支再生,但直到现在我们仍不了解是什么激活了这些基因的表达,因此启动了修复过程。这项研究使得我们朝着有一天能够开发出促进轴突再生的治疗又近了一步。”
轴突是指一些负责发送信号的神经细胞分支。它们通常比负责接收信号的树突更长,更容易受到损伤。
在外周神经系统(脑和脊髓外的神经细胞网络)中,细胞有时会自然再生受损轴突。但在由脑和脊髓构成的中枢神经系统中,受损神经细胞通常不会替换失去的轴突。
Cavalli实验室的博士后研究员Yongcheol Cho博士对实验室培育的外周神经系统细胞进行研究,切断了这些细胞的轴突。他和同事们发现,这会导致钙波沿着轴突向后传送到细胞体。这一钙波是激活轴突损伤机制的一系列反应的第一步。
在外周神经细胞中,HDAC5从细胞核中释放出来是这一链式反应中最重要的一个步骤。研究人员将发现,HDAC5离开细胞核后,开启了许多与再生过程相关的基因。HDAC5还来到了损伤位点帮助生成微管,帮助建立了替代轴突的这一支持结构。
当研究人员对HDAC5基因进行遗传修饰,使得蛋白陷在外周神经细胞的细胞核中时,细胞培养物中的轴突没有再生。科学家们还发现,在细胞培养物和动物中通过定量给予细胞一些药物,使得HDAC5更易于离开细胞核,他们能够促进轴突的再生。
当科学家们在中枢神经系统细胞中寻找相同的链式反应时,他们发现HDAC5从未离开细胞核,且不会去到损伤位点。他们认为无法让这一重要的作用因子离开细胞核,或许是中枢神经系统细胞无法再生出轴突的重要原因之一。
“这给我们带来了希望,如果我们能够找到一些方法操控大脑和脊髓神经元中的这一系统,我们就可以帮助中枢神经系统细胞再生失去的轴突。我们现正致力于这一工作,”Cavalli说。
Cavalli也正与外周神经移植开拓者、整形外科主任、外科学教授Susan Mackinnon博士展开合作。两人都在调查是否能够利用HDAC5或是这一链式反应中的其他元件来帮助恢复神经移植物中的感觉功能。
