本研究的第一个目标是开发并验证一种超高效液相色谱串联质谱(UHPLCMS/MS)方法,在单次运行中量化血小板中合成的TxB2和释放的血清素。第二个目的是将此方法与光透射聚集术(LTA)相结合,选择合适的血小板应答器用于HIT诊断

研究人员通过优化血清素和TxB2同时给药的电喷雾电离和色谱分离。该方法根据欧洲药物管理(EMA)的生物分析方法验证指南进行了验证。采用单克隆抗CD9(克隆ALB6)作为血小板激活剂进行LTA,以筛选良好的应答。

结果显示,本检测采用串联质谱联用正负电喷雾电离交替进行。总运行时间为6分钟。对该方法的标定曲线、精度、准确度、量化下限、结转、选择性和基体效应进行了验证。在供者中,血小板对ALB6的反应差异很大。

本研究开发并验证了UHPLCMS/MS同时定量TxB2和血清素的方法。
