Clinica Chimica Acta:循环游离DNA芯片电泳分析及其与心脏疾病中DNase I的关系

循环游离DNA (cfDNA)与癌症、糖尿病、中风、系统性红斑狼疮、创伤、类风湿关节炎、炎症、感染和心肌梗死(MI)直接相关。本研究从心脏疾病患者血浆中提取cfDNA,检测cfDNA片段的分布,以及cfDNA浓度与双链DNA处理所涉及的脱氧核糖核酸酶I (DNase I)活性酶的关系。

循环游离DNA (cfDNA)与癌症、糖尿病、中风、系统性红斑狼疮、创伤、类风湿关节炎、炎症、感染心肌梗死(MI)直接相关。本研究从心脏疾病患者血浆中提取cfDNA,检测cfDNA片段的分布,以及cfDNA浓度与双链DNA处理所涉及的脱氧核糖核酸酶I (DNase I)活性酶的关系。

研究结果显示,心肌梗死和心绞痛患者的cfDNA浓度明显高于健康对照组。血浆cfDNA的微芯片电泳显示,在一些健康对照受试者中出现单个片段(150–200µbp),在所有心脏病患者样本出现中三个片段(150–200µbp300–400µbp500–600µbp)。 此外,与其他心脏病(胸痛,心绞痛,心房颤动和心力衰竭)患者相比,MI患者中cfDNA比率为150-200µbp / 500-600µbp的比例明显更高。此外,DNase I活性与cfDNA浓度呈正相关。

这些结果表明,心脏病患者的血浆cfDNA可能起源于细胞凋亡,cfDNA150-200 bp / 500-600 bp比率可能是MI的新诊断指标。
