在一项新的研究中,来自美国德州大学西南医学中心儿童医学中心研究所(CRI)的研究人员鉴定出一种新的代谢途径,这种途径在对正常细胞是致命性的条件下协助癌细胞生长繁殖。相关研究结果于2016年4月6日在线发表在Nature期刊上,论文标题为“Reductive carboxylation supports redox homeostasis during anchorage-independent growth”。
 
论文通信作者、德州大学西南医学中心儿科遗传学与代谢部门主任Ralph DeBerardinis博士说,“人们长期认为如果我们能够靶向肿瘤特异性的代谢途径,那么就有可能开发出一种有效地治疗癌症的方法。这项研究发现两种非常不同的代谢过程以细胞适应与癌症恶化相关性应激(stress associated with cancer progression)所特别需要的方式相关联起来。”
 
这项研究揭示出癌细胞利用两种已知的代谢途径---戊糖磷酸途径(PPP)和克雷布斯循环(Krebs cycle, 也称作柠檬酸循环)---的替代途径抵抗毒性分子。这些毒性分子是通过氧化应激杀死细胞的活性氧(ROS)。
 
这项研究建立于DeBerardini博士实验室早前的研究发现:克雷布斯循环,即细胞用来产生能量的一系列化学反应,在某些情形下能够反向循环,从而促进癌细胞生长。
 
DeBerardinis博士说,大多数正常细胞和癌细胞通过附着到富含营养物的胞外基质(ECM)上进行生长。他说,“它们依赖于基质附着来接收促进生长的信号和以一种促进细胞生长、增殖和存活的方式调节它们的代谢。”他补充道,从这种基质上脱落会导致ROS突然增加,这种增加对正常细胞是致命的。癌细胞似乎有一种变通方法。
 
哈佛医学院细胞生物学家Joan Brugge博士2009年发表在Nature期刊上的一项划时代研究(Nature, 3 September 2009, doi:10.1038/nature08268)中,就已报道当从胞外基质上脱落时,健康细胞受到破坏。有趣地,DeBerardinis博士说,那项研究也发现将一种癌基因---一种潜在地导致癌症的基因---导入正常的细胞中,会导致它在行为上类似于癌细胞,而且在从基质上脱落后存活下来。
 
DeBerardinis博士说,“另一项于2015年11月发表在Nature期刊上的研究(Nature, 12 November 2015, doi:10.1038/nature15726)来自CRI主任Sean Morrison博士实验室,发现罕见的能够从原发性肿瘤中脱落下来并且成功地转移到身体其他部分的皮肤癌细胞能够阻止ROS达到危险的高水平。”
 
几十年前,人们就已知道PPP是NADPH的主要来源,其中NADPH提供还原当量(即电子)来清除ROS;然而,PPP在细胞的细胞质中产生NADPH,然而ROS主要是由另一种被称作线粒体的亚细胞结构产生的。
 
DeBerardinis博士说,“如果把ROS比作火,那么NADPH就好似是水,被癌细胞用来浇灭火。” 但是,来自PPP的NADPH如何能够处理细胞中一个完全不同的部分产生的ROS应激?DeBerardinis博士说,“我们所做的就是发现这是如何发生的。”
 
在当前的这项研究中,研究人员证实癌细胞利用一种“搭载(piggybacking)”系统携带来自PPP的电子进入线粒体中。这种电子运动涉及细胞质中发生的一种特殊反应,这种反应类似于克雷布斯循环的一种逆反应,将来自NADPH的电子转移到一种被称作柠檬酸的分子。接着,柠檬酸进入线粒体,激活另一种途径从而导致电子释放同时在产生ROS的位点上产生NADPH,这就允许癌细胞不用附着到基质上也能存活和生长。
 
他说,“我们已知道PPP和克雷布斯循环给癌细胞提供代谢益处。但是我们并不曾知道它们以这种异常的方式存在关联。引人关注的是,正常细胞不能够利用这种机制运输NADPH,因而因较高的ROS水平而死亡。”
 
DeBerardinis博士强调,这些发现是基于体外培养的细胞模型作出的,还必需开展更多的研究来测试这种途径在活的有机体中的作用。他说,“我们特别兴奋地测试这种途径是否是肿瘤转移所必需的,这是因为癌细胞需要在从基质上脱落的状态下在血液循环中存活以便发生转移。”
