NEJM:前列腺癌遗传性DNA修复基因突变谱被揭示,复杂度超出想象

7月6日华盛顿大学的科学家们在《新英格兰医学》(NEJM)上发表了一项前列腺癌研究,认为DNA修复基因的遗传性突变在转移性前列腺癌中相当普遍。这些突变对转移性前列腺癌做出的贡献比人们想象的更为广泛,超过10%的人群,这从另一角度也显示出转移性前列腺癌的复杂性和异质性。 在总计7个系列研究,合计692位转移性前列腺癌患者中,超过10% (82位)的侵袭性前列腺癌男性患者,携带DNA修复基因的遗传性突变。这一比例是普通人的四倍多,是局限性前列腺癌患者的两倍多。有82位(11.8%)至少有一个与DNA修复有关的遗传种系基因突变。在84个突变中,有2位患者同时有2个突变。在84例次突变中,79例次是删除突变,5次是已知的错义突变。 在84例次中,共发现16个基因突变。其中BRCA2 (37人[5.3%]), ATM (11人[1.6%]), CHEK2 (10人 [1.9% of 534 men with data]), BRCA1 (6人 [0.9%]), RAD51D (3人 [0.4%]), and PALB2 (3人 [0.4%])。 上述基因突变表明,一些DNA修复基因的遗传性突变(
