Cell Stem Cell:发动造血干细胞的增殖“引擎”需要一关键分子

近日,来自日本的科学家们发现在骨髓移植等应激情况下,造血干细胞和祖细胞内(HSPC)的p38MAPK能够激活嘌呤代谢推动细胞进入细胞周期促进细胞增殖,从而对应激产生应答反应。该研究加深了人们对于造血应激情况下造血干细胞和祖细胞内代谢活动如何变化以及细胞增殖如何启动的认识。

之前研究表明,在正常情况下造血干细胞通过激活糖酵解等一些特定代谢途径保持休眠状态,但是在骨髓移植等应激造血过程中,这些代谢活动如何变化我们仍然没有一个清晰的认识。

在这项最新研究中,研究人员报告称他们发现p38MAPK家族中的一个成员p38α在小鼠应激造血过程中启动造血干细胞和祖细胞(HSPC)的增殖方面发挥重要作用。研究表明在受到应激后,HSPC中的p38MAPK会立即被磷酸化,促进HSPC进入细胞周期。而条件敲除p38α会导致应激造血过程的损伤,造成HSPC增殖启动被延误。

研究人员还从机制上进行了探讨,他们发现p38α信号途径能够增加HSPC中一种叫做肌苷-5`-单磷酸脱氢酶2(IMPDH2)的分子的表达,导致氨基酸和嘌呤相关代谢物水平发生变化,同时还会在体内和体外情况下下改变细胞周期进展。

综上所述,这项研究揭示了p38α介导的信号途径如何改变HSPC代谢从而产生应激应答促进细胞周期恢复,这对于应用造血干细胞进行血液疾病治疗以及深入了解白血病发病机制找到治疗方法具有重要提示意义。
