PNAS:人体中循环的登革1型病毒具有高度传染性

四种登革热病毒(DENV)是蚊子传播的人类黄病毒。 至今,我们对于DENVs如何与人类宿主相互作用,导致无症状,轻度或严重疾病还知之甚少,部分原因是实验室模型是人类DENV疾病的不良替代品。

病毒学家对在人体中复制的DENVs的特性,与在实验室细胞系中繁殖的病毒的特性之间的区别具有浓厚的兴趣,后者被广泛用于研究和疫苗开发。

最近,研究人员使用来自斯里兰卡DENV 1流行病临床样本和新的超灵敏检测,比较了人类血浆中DENVs和在实验室细胞系中经过一次传代后的病毒的特性。

血浆中的DENV比细胞中生长的病毒感染性高50700倍。与在人体中循环的DENV相比,由实验室细胞系产生的DENV在结构上是不成熟的,并且对人抗体的中和过敏。人血浆和细胞培养物衍生的病毒具有相同的基因组序列,表明这些表型差异是由于血浆病毒的成熟状态决定的。几种登革热疫苗正在开发中。最近的研究表明,在细胞培养测定中中和DENV的疫苗诱导的抗体不足以保护人免受DENV感染

关于人类与细胞系中产生的DENV之间结构差异的结果可能是了解疫苗失败和开发更好的疫苗评估模型的关键。
