Oncogene:NF-ĸB-Activin A信号轴能够增强前列腺癌的转移

研究发现,转录组分析表明了在这两种动物模型中,干细胞特性基因、促炎症细胞因子和TGF-β家族成员Activin A在癌症细胞中的表达均提高。一致的是,临床数据分析阐释了Activin A的表达在转移性癌症中特异性天天搞,并且与分层高风险PCa患者的不良预后有关。更多的是,微环境能够通过激活NF-κB诱导Activin A的表达。Activin A表达水平的提高能够通过激活Smad和ERK1/2信号来增强ALDHhi CSC-like表型的侵入性和PCa的增殖,从而驱使转移的发生。Activin A的抑制或者Activin受体的抑制能够显著的减少斑马鱼和小鼠异种种植模型中CSC-like亚群、侵入、转移性生长和骨病变的形成,表明了NF-κB依赖的Activin A在PCa转移中的功能作用。

最后,研究人员指出,他们的研究阐释了人类PCa细胞能够在斑马鱼和小鼠中与微环境表现出相当响应。因此,结合两种动物模型,研究人员阐释了微环境依赖的actinvin信号是PCa转移的必要驱使因子,同时具有作为治疗靶标的可能。
