Aging: 线粒体多肽humanin可以调节寿命和健康

百岁老人的子女更有可能成为百岁老人,他们血液循环中的humanin水平比年龄匹配的对照对象高得多。

Humanin是线粒体基因组内由短的开放阅读框编码的一个新的多肽家族的成员。它在动物中是保守的,并且具有神经保护和细胞保护作用。

最近,研究人员发现,在秀丽线虫中,humanin的过度表达足以增加寿命,这依赖于daf-16/Foxo信号通路。

Humanin转基因小鼠具有许多表型,与蠕虫表型类似。此外,与外源性humanin处理类似,Humanin转基因小鼠可以增加对毒素干扰的保护。

用强效的humanin类似物HNG对中年小鼠进行每周两次的治疗,可以改善代谢健康参数,降低炎症标志物。

在多个物种中,humanin的水平通常会随着年龄的增长而下降,但在这里研究人员表明,在裸鼹鼠中,humanin的水平出奇的稳定。

此外,百岁老人的子女更有可能成为百岁老人,他们血液循环中的humanin水平比年龄匹配的对照对象高得多。

进一步将humanin与健康寿命联系起来的证据是,研究人员观察到humanin水平在阿尔茨海默病和MELAS(线粒体脑病、乳酸中毒和卒中样发作)等人类疾病中会降低。

这些研究共同证明了humanin与改善健康状况和延长寿命有关。
