CLIN CANCER RES:BMP4诱导M2巨噬细胞极化促进膀胱癌进展

膀胱癌目前是一道临床难题,也是社会性难题。诊断时,大部分病人表现为非肌层浸润肿瘤,复发率高,可以进展为肌层浸润肿瘤并转移。膀胱组织中的骨形成蛋白(BMP)依赖信号通路可以促进尿路上皮细胞分化调节尿路上皮稳态。但是BMP配体在膀胱癌中的作用尚不清楚。CLIN CANCER RES近期发表了一篇文章研究这一问题。

膀胱癌目前是一道临床难题,也是社会性难题。诊断时,大部分病人表现为非肌层浸润肿瘤,复发率高,可以进展为肌层浸润肿瘤并转移。膀胱组织中的骨形成蛋白(BMP)依赖信号通路可以促进尿路上皮细胞分化调节尿路上皮稳态。但是BMP配体在膀胱癌中的作用尚不清楚。CLIN CANCER RES近期发表了一篇文章研究这一问题。

作者分析了68例尿路上皮癌组织和正常组织,分析BMP表达和巨噬细胞标志。在体外实验中通过膀胱癌细胞系和外周血巨噬细胞研究其机制。研究结果发现,BMP4表达与Ⅱ型巨噬细胞分化有关。体外实验表明含有BMP4的培养基中膀胱癌细胞系更倾向于向M2亚型巨噬细胞分化,通过IL10分泌和表达可以观察到。通过一系列膀胱癌标本,作者发现在晚期及未分化的肿瘤中BMP4表达升高且与上皮间质化(EMT)密切相关。肿瘤组织pSmad1,5,8染色表明EMT信号减弱,原因是miR-21表达增加导致BMPR2表达降低。

文章最后认为,膀胱癌细胞BMP4分泌为肿瘤免疫微环境提供了必须的M2信号。此外,miR-21导致的BMPR2下降使得肿瘤细胞分化程度降低,有助于肿瘤生长。
