近日,国际学术权威刊物自然出版集团旗下子刊、神经精神病学顶尖杂志《Molecular Psychiatry》在线发表了南方科技大学生物系刘东课题组的一篇研究论文。

ADHD(注意缺陷多动障碍)是常见的儿童行为障碍,其症状可持续至成年,常与多种精神疾病共患,对个人、家庭和社会造成诸多不良影响。ADHD的核心缺陷是执行抑制功能的神经元出现问题,引起多动、冲动和注意缺陷。我们团队早前提出神经元发育缺陷是ADHD的重要致病原因假说,现通过广泛收集病人样本,对ADHD执行抑制功能的全基因组(汉族人群)数据的详细分析(GWAS)和生物信息学验证,发现MICALL2与人的ADHD症状密切关联,MICALL2调节神经元轴突发育。

作为模式动物,斑马鱼研究已具备较为完善的遗传操作手段。斑马鱼与人类基因同源性强(87%),具有获得大量实验样本,养殖成本低,实验操作简便等优势。通过降低斑马鱼MICALL2同源基因micall2b的表达,我们发现幼鱼一些脑神经元发育异常,引起多动-冲动行为,表明抑制功能缺陷。有趣的是,该异常行为可被治疗ADHD药物缓解/纠正(图一)。因此,MICALL2是一个新ADHD致病基因。

卢青、张樱腊共同完成斑马鱼ADHD疾病模型建立和分析。我们拟运用同样的技术手段筛选ADHD致病候选基因,并建立突变体鱼系。在成功建立ADHD斑马鱼模型基础上,将开展ADHD治疗药物的筛选工作。
