日前,The Scientist杂志评选出了 2013 年度生命科学领域10大最具创新性及竞争力的产品与技术(因有两项并列排名,实为12项)。

1、 nVista HD

Inscopix 开发的微型荧光显微镜nVista HD (mini fluorescence microscope)对神经元功能和神经环路研究意义重大。以往难于检测自由活动的状态动物的神经活动:常规显微镜太大,电生理技术则难兼顾到单细胞活 动和整个脑神经元网络活动这两个层面。nVista HD能真正实现单神经元细胞水平观测、同时实时检测多达数千神经元的活动,从而实现对整个神经网络的观测;nVista HD只有两克重,动物带着它可以自由的活动,从而能完成对特定行为学任务的研究。


2、 X-MAN

传 统报告质粒是在过表达模式下工作的,不能反映生理表达量下的情况。Promega开发的X-MAN报告系统则不同,其细胞株含内在的报告子,不需要使用传 统的报告质粒,该类试剂盒包括两种类型:一种为含NanoLuc luciferase报告子的细胞株,无需高表达,在生理表达量下即可开展检测;另一种为含HaloTag的细胞株,该细胞株应用更广泛:兴趣蛋白与 HaloTag表达后,可以通过添加荧光蛋白检测目标蛋白与其它蛋白的相互作用。


2、SmartFlare RNA 探针

EMD Millipore开发的SmartFlare RNA检测探针(SmartFlare RNA Detection Probes)无需抗体,无需杀死细胞,完全在生理状态下,通过荧光检测内源性RNA。SmartFlare RNA Detection Probes检测以往使用固定的死细胞开展实验的方式不同,是在活细胞环境下开展,具有重要意义:针对靶RNA的寡核苷酸与纳米金颗粒结合,与细胞孵育过 夜,从而经胞吞摄入后,与靶基因RNA分子结合,即可通过流式细胞仪或荧光显微镜检测,而这个外源性分子在几天后可通过胞吐排除,细胞又恢复原有状态。


3、SR GSD 3D

荧 光显微镜的分辨率约为200 nm,即便是高分辨率显微镜,虽然XY平面上分辨率可达20 nm,但在Z轴只能达到600–700 nm的分辨率,不能用于观测三维分子结构。Leica Microsystems的SR GSD 3D成像系统,在XY平面上分辨率为20 nm,同时在Z轴达到70 nm的分辨率。其圆柱形镜头结合软件使这种显微镜能判断出某点与观测平面的相对位置(上还是下),并将相应信息合成为3D影像。利用这种显微镜使细胞结构 显现接近了光镜水平,更助于了解细胞内结构和功能。


4、NanoString

乳 腺癌患者因担心肿瘤复发易导致过度医疗,既浪费金钱,并导致伤害。NanoString开发的乳腺癌预后基因信号芯片(Prosigna Breast Cancer Prognostic Gene Signature Assay)通过检测乳腺癌患者肿瘤细胞基因表达谱,可判断患者肿瘤复发风险。如果患者对内分泌治疗反应良好,通过这种芯片评分就能评估病人是否有必要开 展化疗和放疗。这种芯片同样能判断乳腺癌病理类型,相比临床上常用的几种biomarker,其能提供更多的生物学信息。而且这种技术花费低,不需要在专 业实验室内完成,可直接在临床应用。


5、ClearColi

大 肠杆菌含潜在的病原体LPS,以大肠杆菌为模式生物生产生物制品要面对复杂的产品纯化问题。Lucigen开发的大肠杆菌ClearColi不表达 LPS,而是表达LPS的无毒前体脂IVA。当前,他们正在致力于将这种大肠杆菌商业化,希望开发出能承载质粒的菌株,该型菌株既能像实验室常规使用的大 肠杆菌一样生产生物产品,同时又没有内毒素污染的问题。


6、人性化的细菌荧光素酶

分 子生物学家将细胞内插入生物荧光基因来检测药效和毒性,其中最常用的是Firefly荧光酶。这类技术一个显著的缺点是必须杀死细胞,加入外源性试剂来显 示荧光素酶活性,即指利用此类技术开展药物毒性研究的话,实验不能连续性开展,而只能选在不同的时间点来分别测定。490 BioTech开发的细菌荧光素酶报告系统则不同,它不需要添加外源性物质,不需杀死细胞。490 BioTech已经能够在多种细胞类型内引入这种细菌荧光素酶系统,包括多种人源细胞株,而且可根据客户实验需要来订做相应产品。虽然490 BioTech的细菌荧光素酶荧光强度比Firefly弱,但是其观测的实时性和连续性具有其它实验方法无法比拟的优势。


7、Syn Vivo

CFD 的SynVivo是一种人工合成的、模拟体内血管环境的微流体芯片,是用于模拟体内实际液流和几何学特征的体外模型。SynVivo由20个标准化的芯片 组成,可模拟仓鼠、大鼠和小鼠不同血管的几何学特征。芯片含有可培养细胞的腔隙,可用来研究细胞(如神经元细胞,肝细胞和肿瘤细胞等)与药物、循环分子间 的相互作用。研究者只需将芯片的孔道覆盖fibronectin类物质来模拟体内环境,进而在孔道上培养细胞,让液体在上面流动模拟自然情况下体内微血管 网内的血液,从而在近似体内真实环境中研究所感兴趣细胞的功能。


8、ADCC报告生物法

我 们的皮肤可感知碰撞力,并主动反馈躲避危险。由金属或是硬塑料做成的机器人则没有这种能力。SynTouch LLC开发的NumaTac是由网状聚氨酯泡沫做成的垫子,并在其中加入感受器让机器能感触、并对碰撞力作出反应。这种泡沫材料可以根据要求任意塑性,让 机器臂如人手臂那样能感触危险并及时作出反应,从而避免与硬物碰撞导致损伤,这类材料具有广阔的应用前景。


9、NOMe-Seq试剂盒

在 表观遗传学上,DNA甲基化和核小体排布调节着基因的表达,因此需要分别检测DNA甲基化和相应位置的核小体状况来了解基因的转录状况。Active Motif的新技术NOMe-Seq (Nucleosome Occupancy and Methylome Sequencing)能让研究人员同时检测DNA分子的甲基化和核小体排布情况。NOMe-Seq实验开始时,人工甲基化那些不被包裹在核小体内的、固 定的染色质的GpC残基,然后逆转交联,释放核小体及其上结合的蛋白(例如转录因子),然后DNA经双亚硫酸盐转化(bisulfite conversion),使未甲基化的胞嘧啶转化为尿嘧啶,然后通过DNA测序确定DNA甲基化谱和被GpC甲基化保护的核小体印迹 (footprint)。


10、NOMe-Seq

单 克隆抗体治疗在肿瘤和自身免疫性疾病中应用较广,常使用抗体依赖性细胞介导的细胞毒性作用(ADCC)报告系统评估单克隆抗体治疗的疗效。但很多人对 ADCC报告系统并不满意:效应细胞辨认靶细胞上的抗体并攻击靶细胞,可是因效应细胞的变异性太大,相同的抗体,在不同的时间用该报告系统检测所得结果差 异很大。为了发展可靠的检测方法,Promega开发了新型ADCC报告系统(ADCC Reporter Bioassay):Promega研发人员从永生化细胞系开发人源的工程T细胞作为效应细胞,做出了这一稳健、便捷的实验报告系统,使用该系统评估 ADCC能节省大量时间,提高了工作效率。


10、定量多重内标

二 代测序等技术定量所得结果面临者证实困难等诸多问题。Horizon Diagnostics开发的定量多重内标(Quantitative Multiplex Reference Standard)由30种突变组成,在样品准备时掺入,可用于证实不同实验技术、不同实验室间的重复性和有效性。
