Nano Lett:肿瘤光声成像研究新进展

11月27日,国际学术期刊Nano Letters 在线发表了中国科学技术大学化学与材料科学学院教授梁高林课题组的研究成果,文章标题为Alkaline Phosphatase-Triggered Self-Assembly of Near-Infrared Nanoparticles for the Enhanced Photoacoustic Imaging of Tumors。该文章报道了一种碱性磷酸酶控制的近红外纳米粒子的自组装用于光声成像信号放大的策略,并在动物肿瘤模型上显示了光声成像信号明显放大的效果。

光声技术对浅表肿瘤的成像具有很高的空间分辨率。然而,利用肿瘤高表达的碱性磷酸酶来激活探针用于肿瘤的光声成像并没有文献报道。梁高林课题组合理设计了一种化学结构式为1P的近红外探针,同时提出了一种碱性磷酸酶控制的、肿瘤细胞内自组装近红外纳米粒子用于肿瘤的增强光声成像策略(见下图)。研究发现1P在碱性磷酸酶的作用下生成去磷酸化的产物1,1被肿瘤细胞摄取后自组装形成纳米粒子从而增强肿瘤的光声成像信号。他们与苏州大学合作,发现纳米粒子的形成在体外会使光声成像信号增大6.4倍。体内肿瘤光声成像结果表明,与碱性磷酸酶抑制剂处理的对照组相比,实验组的肿瘤光声成像信号增强了2.3倍。需要指出的是,如果把1P中的Phe-Phe-Tyr(H2PO3)-OH片段换成其他酶切序列,采用这个策略可以发展出更多的“智能”光声成像探针用于精准诊断他们相对应的癌症。

该研究得到国家重点研发计划、国家杰出青年科学基金以及基金委创新研究群体项目的资助。
