Eur Urol:李翀研究组发现膀胱癌干细胞起源的重要遗传学机制

中国科学院生物物理研究所蛋白质科学研究平台抗体工程实验室李翀与中国科学院微生物研究所、深圳大学的研究人员合作,采用单细胞测序技术对肿瘤干细胞进行基因组学层面的探索,并进行了功能性实验验证。

科研人员采用膀胱癌特异性抗体BCMab1与CD44抗体组合,从3份新鲜的膀胱癌组织中分选膀胱癌干细胞(Bladder Cancer Stem Cells)、膀胱癌非干细胞(Bladder Cancer Non-Stem Cells)、膀胱上皮干细胞(Bladder Epithelial Stem Cells)和膀胱上皮非干细胞(Bladder Epithelial Non-Stem Cells),共计59个细胞进行了单细胞MALBAC扩增和全外显子测序。

通过进化分析发现:膀胱癌干细胞起始于膀胱上皮干细胞或膀胱癌非干细胞。对膀胱癌干细胞中发生突变的21个关键基因进行鉴定后发现,有6个基因未曾在膀胱癌中报道(ETS1,GPRC5A,MKL1,PAWR,PITX2,RGS9BP)。ARID1A,GPRC5A和MLL2联合突变可显着增强“膀胱癌非干细胞”转化成“膀胱癌干细胞”能力。

这一研究成果利用单细胞MALBAC扩增技术结合全外显子测序技术详细描绘了膀胱癌干细胞的基因组学概况,实验性证实了“膀胱癌干细胞”起源于“膀胱癌非干细胞”这一科学问题,揭示了人类膀胱癌干细胞起源的遗传学基础,阐明了癌变细胞中关键突变(Driver mutation)如何调控膀胱癌干细胞的自我更新机制,对于人类膀胱癌的防治具有重要意义。
