新型癌症成像技术助力癌症研究突破性进展

【1】Nat Methods:高端成像技术让癌细胞无所遁形

利用高科技的成像方法,来自华盛顿大学的科学家就可以在组织深处清楚地看到早期发育中的癌细胞,这或许比之前利用荧光蛋白来观察癌细胞要更加清晰直观,相关研究发表于国际杂志Nature Methods上。

研究者Lihong Wang表示,通过遗传性地修饰胶质母细胞瘤细胞使其表达BphP1蛋白,我们就可以利用光声层析成像 (Photoacoustic tomography)在组织1厘米深处清楚地观测成百上千个活的癌细胞,光声层析成像技术是一种新型的无损无创生物医学影象技术;本文研究工作中研究人员首次将深度侵入技术、高分辨率的光声成像技术同可逆转的可变开关非荧光的细菌光敏色素相结合进行研究。

研究者指出,蛋白质的遗传编码可以使得我们对组织深处的生物学过程进行成像并且靶向追踪,而蛋白质的光控开关特性也可用作新的成像功能;BphP1蛋白可以感知不用类型的光,同时相应改变其吸收特性,这种特性就可以帮助研究者利用两种类型的光:红光或近红外光来获取癌性组织的成像结果,同时也可以对成像结果进行对比来获取癌细胞高度敏感性及高分辨率的成像结果。

【2】Nature:活体成像捕捉癌细胞的一举一动

在我们展示影片时,当人们看到肿瘤病变如何演化,都惊讶地站了起来。这是一种认知上的改变。

当 Mikala Egeblad 完成第一个活鼠体内肿瘤细胞的活动影片时,她兴奋不已。在那之前,她已经对显微切片上的样本进行了研究。不过在活的动物体内观察细胞则令人产生更为鲜活的感知。“就像你打开显微镜观察活的老鼠,相同的细胞突然间疯狂地动来动去。”美国纽约冷泉港实验室癌症研究员 Egeblad 说道,“它真的改变了我的想法。”

越来越多的癌症研究人员正在寻找机会观察原生环境中的单个肿瘤细胞。在静态组织培养研究中,研究人员不得不推断肿瘤附近的癌细胞和其他细胞可能在做什么以及它们可能会如何相互作用。一种被称为活体成像的方法能够追踪活的动物体内的癌变,可以将这些互动表现出来,并允许生物学家放大显示导致疾病或抵抗治疗的肿瘤中的少数危险细胞。

【3】Cancer Res:新型成像技术助力癌症研究治疗

刊登在过国际杂志Cancer Research上的一篇研究论文中,来自达特茅斯Geisel医学院的研究人员开发了一种新型的荧光成像技术,其可以不在活组织检查的情况下精确鉴别出靶向癌症疗法的特殊受体。

研究者Kimberley S. Samkoe教授说道,蛋白质的过度表达往往是特殊癌症的一个标志,而且也常常在临床肿瘤学领域通过检测肿瘤来用于开发癌症患者的个体化疗法;蛋白质的表达可以通过对肿瘤组织的总蛋白分析测得,而本文中新型技术的开发可以帮助研究者在不进行侵入性活检的情况下精确鉴别出蛋白质受体的含量。

研究者开发的这种双重追踪体内受体浓度成像(RCI)技术包括同时注射靶向和非靶向的成像制剂,随后研究者对5种肿瘤组织的蛋白表达进行了研究,将RCI测得的数据同临床免疫组化所的的数据进行比较,结果显示,通过RCI测得的蛋白质表达和组织分析所得到的结果具有较强的关联性,常用于测定蛋白质表达的技术,比如蛋白印迹或流式细胞计数等,其和RCI值并无关联性。

【4】JACS:双色“条形码”或可用于机体癌症的精确成像

近日,来自新加坡A*STAR研究所的研究人员对扮演微型二元“条形码”的杆状样单晶体进行分析,揭示了其追踪细胞的特殊功能及防伪措施,相关研究刊登于国际杂志Journal of the American Chemical Society上。

所谓的稀土元素掺杂的增频转换材料具有高潜力的应用价值,比如其可以用于抵御犯罪的发生,亦可抵御癌症的发生,然而截止到目前由其制成的单晶纳米晶体并不实用,因为其尺寸太小而不足以使得研究人员用常规的光学显微镜进行观察。

这项研究中,研究人员Xiaogang Liu就通过合成不同颜色的微尺度杆(微型杆)克服了上述问题,研究者开发的这种多色彩微尺度杆含有红色、绿色和蓝色条,这些不同颜色的棒状条是由稀土元素掺杂的增频转换材料制成,即NaTF4,这些不同颜色的棒状条就可以通过标准的显微镜进行观察分析。

文章中,研究者首先通过改变掺杂稀土元素钆的浓度来控制NaTF4的长度,随后通过改变稀土元素镱和铒离子的浓度来调节NaTF4的颜色进而制成微型条形码,研究者Liu表示,这些制成的微型条形码可以产生两种透明的安全油墨,即包含绿色中心和红色指示条的微型杆和包含绿色微型杆的控制油墨,其可以用于加强防伪措施;当利用常规显微镜进行分析时,如果处于低倍镜下时两种安全油墨产生的指纹型并不易于区分,而提高观察倍数就会使得非控制油墨中的红色指示条清楚可见,从而就将两种防伪油墨轻松地进行了区分。

【5】Cancer Res:新型成像技术可有效检测出恶性耐药性的癌症

近日,刊登在国际杂志Cancer Research上的一项研究报告中,来自曼彻斯特大学等处的科学家通过研究开发了一种新型成像检测技术,其可以在肿瘤扩散之前帮助医生们鉴别出更多危险的肿瘤,并且指导临床治疗;文章中研究者详细描述了这种磁共振成像技术如何绘制出缺氧肿瘤存在的区域。

缺氧状态是癌症恶性发展的一个标志,其也会促进肿瘤内部血管的生长,从而促进癌细胞向机体其它部位扩散;这项研究或可帮助开发有效的放疗技术来增强X射线的剂量来有效作用缺氧区域的肿瘤组织,同时也为监测是否放疗或者其它药物有效提供思路。文章中研究者利用一种新出现的名为氧增强的磁共振成像技术来通过将癌细胞植入小鼠机体中,从而绘制出缺氧性肿瘤的位置,该技术未来或可用于癌症病人的临床研究中。

【6】Ann Surg Oncol:新的成像剂可以更好的帮助检测癌症

近日,美国加州大学圣地亚哥医学院的研究人员已经证实由加州大学San Diego Moores癌症中心设计和开发的一种新的成像染料是一种检测和映射已经转移到达淋巴结癌症的有效试剂。

放射性染料叫Tilmanocept,能成功地确定癌变的淋巴结,比现行标准染料更好地标记癌症。 III期临床试验的结果在线发表在Annals of Surgical Oncology杂志上。

发明者David R. Vera博士说:Tilmanocept是一种专门检测淋巴结的新型放射性试剂。加州大学圣地亚哥分校医学院所开发的这一试剂为外科医生提供了新的工具,以准确检测黑色素瘤和乳腺癌。在2013年3月13日,Tilmanocept获得FDA批准。

癌症诊断后,医生希望可以肯定这种疾病还没有扩散到患者的淋巴结,尤其是前哨淋巴结。

【7】Sci Transl Med:新的成像软件能发现癌症

研究人员创建了一种叫做“C-路径”的电脑程序,这种程序可对乳腺组织做显微图象的扫描以寻找6000种以上的癌症特征。

该软件在2组妇女中帮助预测了乳腺癌的严重性,它可能是一种判断某位患者存活机会的有用工具。

自1920年代以来,病理学家大多依赖于同一组少数特征来发现组织样本中的异常。 Andrew Beck及其同事研发的C-路径旨在发现可帮助更为精确地反映患者存活结果的癌组织的额外特征。 他们对采自荷兰的一组病人的组织样本做了C-路径的测试。

该软件发现了与不良存活机会有关的一组崭新的特征。

在另外一组来自温哥华的病人中,C-路径根据一套已知的综合性特征及新的癌组织特症预测了这些妇女生存的机会。将组织分类为上皮或基质组织是癌症诊断的一个重要部分,但它需要作更多一点的工作:该研究小组必须教该电脑程序如何用手工标记的样本来发现每种组织的类型。一则相关的《观点栏目》称赞C-路径为第一个潜在可用的电脑化病理系统,但它也指出该软件存在可能阻止其立刻用于医疗机构中的显著的局限性。

【8】ACS Nano:借助纳米颗粒可实现肝癌细胞成像

在多数的恶性肝脏肿瘤的治疗中,手术切除都是第一线的治疗方案。在肝脏肿瘤切除手术中,如果能更精细地区分肿瘤和正常组织的边缘,以及能够观测到微观损伤的区域,对于成功的肿瘤切除手术非常重要。美国纽约纪念斯隆-凯特琳癌症中心的Moritz F. Kircher博士领导的课题组,合成了一种硅包被、表面增强拉曼散射的纳米颗粒(NPs),可以用于肝脏肿瘤成像。

在临床治疗中,恶性肝脏肿瘤的最有效的方法就是手术切除,然而手术切除常常无法全部切除所有的恶性组织。在正常组织和恶性肿瘤组织的边缘非常难以区分,小块的肿瘤组织的遗存会严重影响术后的恢复。随着腹腔镜检查技术和机器手臂手术操作越来越普及,对于正常组织和肿瘤组织的边缘区分的意义更加重要。虽然现在存在很多种肝脏表明成像的工具,比如核磁共振成像,计算机断层扫描,正电子放射断层造影术,以及超声成像等,都存在着这样或者那样的不足。

【9】Nature:开发出新型成像模型 或揭示胰腺癌治疗新靶点

胰腺导管腺癌(pancreatic ductal adenocarcinoma)是一种常见的胰腺癌,其具有极大的致死性,患者的5年生存率仅为6%,目前化疗方法并不能够有效治疗胰腺导管腺癌,部分原因是癌细胞对当前的疗法体系具有较高的耐药性。

近日刊登在国际著名杂志Nature上的一项研究报告中,来自加州大学圣地亚哥医学院等机构的研究人员通过研究开发了一种新型模型,该模型不仅可以帮助研究者追踪体内的癌细胞耐药性,还帮助揭示了一种新型的治疗靶点,早期检测结果或许就可提供一种新型策略来遏制胰腺癌细胞生长。

研究者表示,我们开发的这种新型“报道子”小鼠模型可以在活体动物机体中对干细胞信号进行非侵入性的基于图像的追踪;而利用这种策略,研究人员就发现,干细胞基因Musashi (Msi)是胰腺癌进展过程中的关键元件,尤其是Msi基因的表达水平会随着癌症进展而升高,表达Msi的细胞是驱动癌细胞生长、药物耐受性及患者致死的主要原因。

在确定Msi基因可以促进疾病恶化后,研究人员就开发了一种抵御Msi的新一代反义寡核苷酸抑制剂,这些抑制剂可以有效靶向作用并阻断表达Msi的细胞,从而抑制动物模型机体中的肿瘤生长以及病人机体中的癌细胞,病人机体中的癌细胞往往具有较复杂的突变,而且药物耐受性上具有均一性。

【10】JNM:有望诊治癌症的切伦科夫冷光成像技术

美国核医学学会7月1日表示,新出版的《核医学杂志》报道了名为切伦科夫冷光成像(Cerenkov luminescence imaging)的新型光学成像技术。据文章作者介绍,新技术有望帮助人们诊治癌症和其他疾病,以及更快和更有效地开发放射性药物。

研究负责人、斯隆-凯特灵纪念癌症中心教授简·格林姆博士表示,新型多通道显影剂和技术属于医学成像科学领域的研究前沿,它可能为新的光学成像进入临床应用开辟新途径。格林姆小组认为,自己的研究属于那些首次探讨将切伦科夫辐射应用于医学成像的工作。据悉,加州大学和斯坦福大学科学家参与了研究。

当光在水中传播时,其速度会减慢。而此时速度超过光速的粒子如同突破声障的音爆,会产生 “震波”(或“冲击波”)发出蓝色可见光,该现象被称为切伦科夫辐射。

光学成像是一种分子成像过程。在此过程中,设计出来用于附着在特殊细胞和分子上的发光分子被注入人体血液中,并可为光学成像仪探测到。通常,为便于光学成像仪工作,这些发光分子需要通过体外光源或生物手段进行激活。
