由于突变在老年人中相对较为普遍(超过10%的70岁以上的人口至少存在这些突变之一),根据研究人员的说法,筛选血细胞突变,识别冠心病风险增加的人群、通过生活方式改变减少这些人的风险或治疗干预措施可能具有重大的临床影响。

“未来有更多的工作要做,但这些结果表明,血液细胞前恶性突变是心血管疾病的主要原因,将来可能会根据这些发现使用标准疗法或新的治疗策略进行治疗。”新研究的共同资深作者本杰明·埃伯特(Benjamin Ebert),布拉德(Broad)的研究所成员,哈佛医学院的医学教授和布里格姆妇女医院的血液学家说道。

虽然人类遗传学数据显示CHIP和冠心病之间有很强的联系,但该团队希望揭示潜在的生物学联系。使用易于发展动脉粥样硬化的小鼠模型,科学家们表明,骨髓细胞中一种CHIP突变基因Tet2的丧失导致血管中较大的动脉粥样硬化斑块,证明该突变可加速小鼠的动脉粥样硬化。

据他们认为,动脉粥样硬化是一种慢性炎症的疾病,可能由于血管壁中过量的胆固醇而产生。为了在细胞水平上检查这一点,该团队转向了对巨噬细胞的研究,一种可以从CHIP干细胞发育并携带相同突变的动脉粥样硬化斑块中发现的免疫细胞。由于Tet2和其他与CHIP相关的突变已知是可以改变其他基因活性的所谓“表观遗传调节因子”,所以该小组检查了来自小鼠的Tet2突变的巨噬细胞中的基因表达水平。他们发现,突变的细胞似乎是具有促进动脉粥样硬化的炎性分子表达增加的“高度炎症”。为了支持这一发现,具有TET2突变的人在其血液中也具有较高水平的这些分子之一——IL-8。

这项工作表明,CHIP与人类冠心病相关联,CHIP相关基因Tet2的突变导致小鼠动脉粥样硬化,炎症机制可能是该过程的基础。需要更多的工作来显示在CHIP中突变的其他基因是否也导致炎症增加。该小组还正在探索诸如降胆固醇治疗或抗炎药物的干预措施是否可能对CHIP患者有好处等问题。

除了心血管疾病,如自身免疫性疾病和神经变性疾病外,炎症也被认为是调节其他几种衰老情况的疾病。由于CHIP随着年龄的增长频率增加,改变炎症过程的体细胞突变可能会影响多种衰老性疾病,尽管需要更多的工作来测试这种可能性。
