近日,来自佛罗里达大学的一项新的研究显示,机体在运动时会释放一种激素——鸢尾素,这种激素不仅会帮助机体减掉脂肪还有助于预防脂肪的形成,该研究结果已发表于 American Journal of Physiology - Endocrinology and Metabolism。研究人员从25女性捐献者捐献的新鲜的皮下白色脂肪组织(scWAT)中获得人体脂肪细胞,以用来检验鸢尾素褐色脂肪和线粒体呼吸

近日,来自佛罗里达大学的一项新的研究显示,机体在运动时会释放一种激素——鸢尾素,这种激素不仅会帮助机体减掉脂肪还有助于预防脂肪的形成,该研究结果已发表于 American Journal of Physiology - Endocrinology and Metabolism。

研究人员从25女性捐献者捐献的新鲜的皮下白色脂肪组织(scWAT)中获得人体脂肪细胞,以用来检验鸢尾素褐色脂肪和线粒体呼吸作用的影响,前脂肪细胞则是用来研究鸢尾素对脂肪生成和骨生成的影响。培养scWAT的碎片用于探究信号转导通路以及解偶联蛋白1(UCP1)。scWAT中机体对鸢尾素的反应与棕色/米色基因的基础表达水平相关联。

结果发现鸢尾素上调了褐变相关基因的表达,且上调了UCP1蛋白在原代成熟脂肪细胞和新鲜脂肪组织中的表达水平。此外,通过提高细胞能量代谢,鸢尾素还显著增加了细胞产热作用(5 nM)。使用鸢尾素来处理人类scWAT可激活ERK和p38 MAPK信号通路从而增加UCP1的表达。使用特异性抑制剂阻断任一通路都会消除鸢尾素诱导的UCP1表达上调的作用。相反,人类棕色脂肪组织(BAT)中的UCP1对鸢尾素反应并不敏感。棕色/米色基因和FNDC5基因的基础水平与scWAT对鸢尾素的褐色反应呈正相关。此外,鸢尾素可显著抑制脂肪细胞的分化,促进成骨的分化。

总而言之,该研究发现表明,鸢尾素可促进成熟的脂肪细胞和scWAT “褐变”和,并可增强脂肪细胞产热,除此之外,在谱系特异性分化过程中还可抑制脂肪细胞分化促进骨分化。该研究为进一步探索治疗鸢尾素治疗肥胖以及运动相关的骨形成提供了有价值的见解。
