J Endod:根管内二甲双胍通过抑制诱导型一氧化氮合酶表达以及单核细胞募集促进根尖周炎愈合

作者之前的研究显示,根管内二甲双胍能够部分通过调节成骨细胞凋亡改善根尖周炎的治疗效果。但二甲双胍在根尖周炎发展过程中对其他类型细胞的作用还需进一步研究。因此,这篇研究的目的是为了评估它对诱导型一氧化氮合酶(iNOS)表达以及单核细胞募集的作用是否有助于根尖周炎的愈合效果。

作者之前的研究显示,根管内二甲双胍能够部分通过调节成骨细胞凋亡改善根尖周炎的治疗效果。但二甲双胍在根尖周炎发展过程中对其他类型细胞的作用还需进一步研究。因此,这篇研究的目的是为了评估它对诱导型一氧化氮合酶(iNOS)表达以及单核细胞募集的作用是否有助于根尖周炎的愈合效果。

研究使用LPS诱导人单核细胞系Mono-Mac-6内iNOS的表达,并通过western blot检测。检测培养基中亚硝酸盐的含量以定量分析一氧化氮(NO)水平。采用酶联免疫吸附测定检测C-C motif chemokine ligand-2 (CCL-2)的合成。对大鼠实验性根尖周炎进行清创并施以根管内二甲双胍封药与否。通过常规的X线影像以及micro-CT影像评估损伤的进程。通过免疫组化评估iNOS的细胞内表达水平以及单核细胞/巨噬细胞的数量。

结果显示,二甲双胍通过单核细胞抑制LPS诱导的iNOS和NO水平。更重要的是,二甲双胍通过调节iNOS/NO通路抑制LPS促进的CCL-2合成。根管内二甲双胍减小根尖周炎引起的骨吸收并抑制iNOS的表达和单核细胞募集。

结论:这些结果证实了根管内二甲双胍对根尖周炎的治疗效果。通过调节iNOS表达以及NO水平抑制单核细胞募集是二甲双胍的主要作用机制。
