ACS 2015:新型多肽探针有助于检测全身血栓部位

众所周知,机体若出现血栓则非常危险,甚至会致命。所以,越早清除血栓,它能引起机体出现心脏病、中风或其他致命性疾病的风险就越小。若一个人因机体血栓的出现而导致中风,则其再次患中风的风险增加,但是因为血栓位置不同,因此治疗方案也各不相同。为了清除机体的血栓,一些患者需要进行手术,而另一些患者需要进行溶栓治疗。

因此,医生必须精确地找到血栓所在的位置并采取正确的治疗方案。但是现有的医疗条件仅允许他们通过某些扫描检测身体的部分部位。例如,患者往往要进行三种不同类型的扫描:超声检查颈动脉或腿部,磁共振成像(MRI)检查心脏,计算机断层扫描(CT)检查肺部情况。

MGH化学副教授和哈佛医学院放射学教授的Peter Caravan认为,这样无异于盲人摸象。为了检测到血栓的正确位置,病人往往要进行多次扫描,因为若有一种扫描方式,只需扫描一次便可得知病人血栓的位置,这样最是方便省时不过了。Caravan 教授和他的研究团队发现了一种存在于血液中且不溶的多肽探针,且这种多肽与纤维蛋白特异性结合,可以帮助寻找机体的血栓部位。

为了完成实验,研究人员采用了不同的放射性核素、多肽和引入方式。

并将放射性核素(放射性核素显像:将放射性药物引入体内后,以脏器内、外或正常组织与病变之间对放射性药物摄取的差别为基础,利用显像仪器获得脏器或病变的影像)引入多肽,通过PET对动物进行功能性显像。

通过实验,研究人员确定了15个血栓可能存在的部位。经过分析多肽与纤维蛋白结合的程度后,研究人员将其应用于小鼠体内进行检测。但是结果表明,实验室的试管结果与小鼠体内的结果差异明显。

Caravan教授认为,多肽结合纤维蛋白的作用相似,但或许因为小鼠体内的新陈代谢使得某些多肽无法发挥作用。因此还需要更进一步的研究以找到更科学和稳定的方法。
