ANN RHEUM DIS:端粒长度与痛风患者炎症频率和罹患心血管疾病的相关性

痛风患者比健康人的端粒长度短,反映了增加的细胞衰老。端粒缩短与痛风患者的发作次数和罹患心血管疾病相关。

慢性炎症与衰老相关,而衰老是公认的血管疾病强有力的预测指标,近日,风湿领域权威杂志Annals of the Rheumatic Diseases上发表了一篇研究文章,该研究旨在验证炎症加速衰老,也会同时增加痛风患者罹患血管疾病风险的假设。

研究人员通过量化端粒长度(TL)来评价细胞复制性衰老,共纳入145名荷兰痛风患者和273名健康人,并在新西兰474名痛风患者和293名健康人中进行结果验证。研究测量CD4 +、CD8 + T淋巴细胞,B淋巴细胞,单核细胞,自然杀伤细胞和浆细胞样树突状细胞的TL。 此外还对TL和端粒酶活性的潜在暂时性差异进行了评估。

研究人员发现健康人外周血单核细胞的TL随时间而下降,反映了正常衰老过程。而痛风患者的端粒长度更短(P=0.001,R2=0.01873)。事实上任何年龄痛风患者端粒的侵蚀程度均比健康人高(P<0.0001,R2=0.02847)。有心血管疾病的痛风患者端粒最短。TL是痛风患者罹患心血管疾病的独立危险因素(P=0.001)。TL与痛风发作次数呈负相关(P=0.005)。

由此可见,痛风患者比健康人的端粒长度短,反映了增加的细胞衰老。端粒缩短与痛风患者的发作次数和罹患心血管疾病相关。
