在这项研究中,我们开发了一种含有淫羊藿苷的生物活性复合支架,可以提供结构和机械支持,促进骨再生,从而在建立的SAON兔模型中填充CD手术后的骨缺损。创新使用低温3D打印技术来制造聚(乳酸-乙醇酸共聚物)/β-磷酸钙/淫羊藿苷(PLGA/TCP/淫羊藿甙,PTI)支架。体外测试了PTI支架的细胞相容性,并且在SAON兔模型中体内评估了PTI支架的成骨特性。

结果表明,打印的PTI支架具有理想的促成骨仿生结构,可为骨再生支架提供较好的机械支持和稳定的淫羊藿苷的释放。此外,体内研究表明,与PLGA/TCP(PT)支架相比,PTI支架可更好的增强新生骨组织的力学性能,并改善SAON兔模型植入区内的血管生成。体外研究中,使用MC3T3-E1细胞探究成骨细胞的成骨机制,发现淫羊藿苷可以促进MC3T3-E1细胞向内生长并调节其进入成骨细胞分化。与PT支架相比,PTI支架具有更好的生物降解性、生物相容性和成骨能力。

总之,该研究结果表明,掺入生物活性植物化合物的PTI复合支架可用于难治性SAON患者的骨缺损治疗。
