Nat Immunol :马凤霞/李程/罗鸿博合作发表中性粒细胞稳态和炎症状态下的单细胞发育和异质性图谱

中性粒细胞是天然免疫系统的主要组成部分,作为循环白细胞中数量最多的细胞类型,是宿主抵御入侵细菌等病原体的第一道防线。然而,目前对中性粒细胞的分群主要是依靠经典的形态学,中性粒细胞的异质性一直存在争议。

中性粒细胞是天然免疫系统的主要组成部分,作为循环白细胞中数量最多的细胞类型,是宿主抵御入侵细菌等病原体的第一道防线。然而,目前对中性粒细胞的分群主要是依靠经典的形态学,中性粒细胞的异质性一直存在争议,没有统一的认知,这种分歧阻碍了中性粒细胞研究的发展。与其他血液和免疫细胞类型相比,中性粒细胞中基因表达的数量和水平较低,从技术层面上对RNA的捕获更加困难。另外,由于中性粒细胞的半衰期只有15-20小时,对实验操作细节也提出了更高的要求。

2020年7月27日,中国医学科学院血液病医院(中国医学科学院血液学研究所)马凤霞课题组、北京大学生命科学学院李程研究组、哈佛大学医学院罗鸿博研究组合作在Nature Immunology杂志上发表了题为“Single-cell transcriptome profiling reveals neutrophil heterogeneity in homeostasis and infection”的研究论文。

作者们通过优化实验条件,分别对小鼠稳态下的骨髓、外周血和脾脏进行中性粒细胞分离和单细胞RNA测序,并结合生物信息学方法建立了稳态条件下的中性粒单细胞图谱。按照发育成熟的顺序,骨髓中的中性粒细胞可以分为5群,包括3个处于分裂阶段的亚群(G0、G1、G2) 和2个分裂后的较成熟亚群(G3、G4)。作者们针对颗粒蛋白、细胞周期、杀菌功能等方面,详细解析了每个群体的特征,并结合流式细胞标记等方法验证了这些群体的存在。研究发现,虽然单细胞RNA测序定义的异质性细胞群体与经典形态学分群整体具有较高相关性,但是单细胞测序技术能辨别出形态学相同、而转录组有差异的G3和G4粒细胞群体,增加了对骨髓成熟粒细胞异质性的认识。根据颗粒蛋白基因在亚群之间表达的异质性,作者们提出了颗粒蛋白产生的“分选假说”(sorting mechanism),并通过转录调控网络分析鉴定出了中性粒细胞发育过程中特异上调的新转录因子。

作者们接下来分析了外周血及脾脏中的中性粒细胞,发现外周器官中主要存在3群具有不同转录组特征的成熟中性粒细胞PMNa、PMNb和PMNc。RNA速度(RNA velocity)分析揭示了这些成熟群体有不同的骨髓来源和复杂的群体间相互转化关系:1)PMNa和PMNb主要分别由骨髓的G3和G4发育而来;2)PMNa和PMNb均可发育成PMNc,但是PMNa和PMNb之间很少有转化发生。利用免疫荧光成像技术,作者们进一步验证了高表达干扰素刺激相关基因的成熟中性粒细胞(PMNb)在脾脏中独特的空间分布,提示该亚群可能具有不同于其他亚群的宿主防御功能。此外,作者们还将之前文献中使用单细胞测序定义的异质性群体映射到本研究建立的中性粒细胞参考图谱,统一了领域对小鼠稳态下中性粒细胞异质性的分类。

最后,为了研究细菌感染对中性粒细胞发育及亚群的影响,作者们分离了急性腹膜炎小鼠的骨髓、外周血、脾脏及感染部位的中性粒细胞进行单细胞RNA测序和分析。发现在炎症状态下的中性粒细胞的异质性群体没有改变,但各群体的数目比例、功能特征、转录因子表达和群体转化路径等发生了明显变化。分析和实验结果共同表明,急性髓系造血主要通过增强早期中性粒细胞分裂活动和缩短分裂后中性粒细胞成熟时间来共同完成,这些发现为急性细菌性感染治疗提供了潜在分子靶标。

总的来说,该研究首次从单细胞尺度系统性地描绘了中性粒细胞在稳态和炎症状态下成熟、分化和命运决定过程中的异质性群体和转录组动态变化,整合了领域内已有的关于中性粒细胞异质性分类,为进一步研究中性粒细胞提供了有价值的参考数据资源。
