Kidney Int:增强干细胞在缺血再灌注肾损伤中功效的新方法!

研究表明,间充质干细胞(MSCs)对实验性缺血再灌注急性肾损伤(IRI-AKI)的治疗具有一定的效果。其中,免疫调节是MSCs治疗的重要机制之一。白细胞介素-17A(IL-17A)预处理的MSsC更具免疫抑制性,而体外免疫原性的变化很小。近期,一项发表在杂志Kidney Int上的研究旨在评估给予IL-17A-预处理的MSCs导致IRI-AKI小鼠的急性肾小管坏死评分,血清肌酐以及BUN是否显着低

研究表明,间充质干细胞(MSCs)对实验性缺血再灌注急性肾损伤(IRI-AKI)的治疗具有一定的效果。其中,免疫调节是MSCs治疗的重要机制之一。白细胞介素-17A(IL-17A)预处理的MSsC更具免疫抑制性,而体外免疫原性的变化很小。近期,一项发表在杂志Kidney Int上的研究旨在评估给予IL-17A-预处理的MSCs导致IRI-AKI小鼠的急性肾小管坏死评分,血清肌酐以及BUN是否显着低于给予MSCs。此项研究结果显示:在共培养的脾细胞中,IL-17A预处理的MSCs显着增加了CD4 + Foxp3 + Tregs的百分比,降低了concanavalin A诱导的T细胞增殖。此外,与接受MSCs治疗的小鼠相比,接受IL-17A预处理的MSCs治疗的IRI-AKI小鼠的血清IL-6、TNF-α和IFN-γ水平显着降低,而血清IL-10水平更高,脾脏和肾脏Treg的百分比也更高。由PC61(抗-CD25抗体)对Treg的消耗逆转了IL-17A-预处理的MSCs对IRI-AKI小鼠增强的治疗功效。另外,IL-17A还上调了COX-2的表达并增加了PGE2的产生。塞来昔布阻断COX-
