这项研究的发现,如果适用于人类的话,将会对血细胞移植和使用血细胞的临床方法和研究方法(如基因治疗或基因编辑)产生影响。

这项研究将研究进一步推动血液再生疗法的开发,但是这些研究人员认为它也适用于各种细胞,并将产生关于病变的或受损的组织再生的新见解。

论文第一作者、哈佛干细胞研究所的Alejo Rodriguez Fraticelli博士说,“我们的研究结果表明,当不将造血干细胞和它们的多能性不那么强的后代---血祖细胞---从它们的天然环境中移除的情形下以及当在实验室中或在移植期间开展研究时,它们表现得有点不同:这会导致它们产生的血细胞谱系存在差异。”

由于缺乏适当的工具来研究血液在身体的天然环境中是如何形成的,大多数关于单个血细胞来自何处的研究都是在移植后开展的。在这种情况下,移植的细胞将会受到“扰乱”,或者离开了它们的天然环境。根据这些研究人员的说法,当前的血细胞形成模型更可能代表着造血干细胞的天然后代的细胞谱系潜能的路线图。

对Rodriguez Fraticelli来说,这突出了在天然环境下研究血液再生的重要性。他说:“展望未来,我们需要开发出一些新的方法来更好地预测哪类细胞最适合用于治疗,例如用于对细胞进行重编程和编辑。”

在这项新的研究中,这些研究人员利用转座子对造血干细胞和它们的祖细胞后代进行标记,随后在天然的不受干扰的血液再生过程中追踪它们。转座子是当遇到转座酶时能够跳跃到DNA的随机位点上的一段遗传密码。

这项研究为天然环境下的正常血液再生或血液产生的路线图进行大幅修订提供了证据,并强调在这些条件下造血干细胞和它们的祖细胞后代如何展现出独特的性质。
