近日,来自美国阿拉巴马大学的研究人员在国际学术期刊Cell Reports上发表了一项最新研究进展,他们找到利用化合物促进体细胞重编程获得诱导多能干细胞的新方法。

虽然人体内200多种不同的细胞类型具有相同的基因组,但是每种细胞类型都存在独特的基因表达程序。在体细胞重编程获得诱导多能干细胞的过程中,清除原始细胞的体细胞转录程序是一个关键事件。

在这项研究中,研究人员找到了一种新策略,他们利用小分子化合物干扰体细胞的转录记忆帮助体细胞重编程获得多能干性。研究人员发现靶向BET家族蛋白乙酰赖氨酸结合口袋的小分子化合物能够大大增强重编程过程。更进一步研究表明小分子化合物靶向BET的bromodomains能够下调或关闭成纤维细胞的体细胞基因表达,还会在重编程早期阶段导致成纤维细胞形态丧失。

综上所述,研究人员通过实验证明借助靶向BET bromodomains的化合物清除体细胞的转录记忆可以促进细胞发生命运转变。该研究为提高诱导多能干细胞的得率提供了一个新策略。


据一篇发表在国际学术期刊Human Gene Therapy上的综述文章介绍,将治疗性干细胞直接移植到中枢神经系统是治疗溶酶体贮积症(lysosomal storage diseases)造成的神经疾病的一个非常具有前景的新方法,不过将基于LSD动物模型开发的干细胞治疗方法转化为用于人类疾病治疗的有效方法仍然面临着许多重大挑战。

溶酶体贮积症(LSD)是一组遗传性代谢疾病,是由于基因突变致溶酶体中有关酸性水解酶缺陷,导致机体中相应的生物大分子不能正常降解而在溶酶体中贮积,引起细胞组织器官功能的障碍。目前已知溶酶体贮积症至少包含50种不同的单基因遗传病。

文章共同作者Faez Siddiqi和John Wolfe在这篇综述文章中对可以用于治疗神经疾病的不同干细胞类型进行了讨论。他们回顾总结了一些最新的研究进展,描述了科学家们在神经干细胞工程改造以及如何将干细胞导入中枢神经系统,使得干细胞对LSD病人的治疗能力达到最优化方面做出的努力。


来自美国塔夫茨大学的研究人员开发了一种获得具有快速分化能力的人类神经干细胞的新技术,获得的神经干细胞可以用于包括人类大脑三维模型构建在内的多种组织工程学研究,相关研究结果发表在国际学术期刊Stem Cell Reports上。

根据文章介绍,研究人员可以把人类成纤维细胞和脂肪来源干细胞转变成稳定的人类诱导神经干细胞系,这些干细胞可以在最短四天内就获得活跃神经元的特征,通常这一分化过程需要大约四个星期。获得的神经干细胞可以被冻存,无限传代,同时还具有一些独特属性让它们能够在体外与其他细胞类型一起生长。研究人员在鸡胚胎早期阶段注入人类诱导神经干细胞,发现这些细胞可以融入中枢神经系统和外周神经系统。

这并非第一次获得人类诱导神经干细胞的研究,但是这项研究让这一过程变得更加简单快速,相比于现有的操作流程也更加可靠。


近日,一项刊登在国际杂志Science Advances上的研究报告中,来自加利福尼亚大学的研究人员通过研究发现了一种简便且有效的方法,即给诱导人类多能干细胞“喂以”机体中的天然分子腺甘酸,就可以使其再生成为骨质组织,这种源于干细胞的骨质组织可以在不产生肿瘤或诱发感染的情况下帮助修复小鼠的颅骨缺陷。

该项研究或将帮助研究者开发治疗严重骨质缺失及遭受外伤性骨创伤患者的新型再生医学疗法,同时研究者或许也能够开发出一种低廉方便的方法制造纯种的成骨细胞。研究者Shyni Varghese教授表示,我们的一个研究目的就是通过开发有效简便的方法来对人类细胞和组织进行工程化操作,从而开发新型的再生疗法,诱导多能干细胞能够转变为机体中任何一种类型的细胞,比如肌肉细胞、心脏细胞等,这个过程称之为分化,正由于具有这种特性,因此诱导多能干细胞才具有一定的治疗潜力,帮助修复或再生多种组织和器官。


过去几年发表的计算研究已提出癌症的不同主要风险因素,从干细胞的随机突变到环境致癌物。在一项新的研究中,来自美国田纳西州孟菲斯市圣裘德儿童研究医院和英国剑桥大学的研究人员在小鼠体内测试了这些理论,发现干细胞中发生的突变确实发挥着一种显著性的作用。相关研究结果发表在2016年8月25日那期Cell期刊上,论文标题为“Multi-organ Mapping of Cancer Risk”。

论文共同通信作者、剑桥大学英国癌症研究中心主任Richard Gilbertson说,“一种观点在科学界流行了好几年---一些人说癌症是‘坏运气’,这是因为干细胞中的突变是偶然产生的,然而其他人声称环境致癌物在癌症产生中发挥更加重要的作用。这种不一致性主要来自于使用不同的数学模型来研究现存的人类癌症和干细胞数据,因此很难梳理出单个因素的影响。因此,我们在真实的实验模型中测试这些不同的观点,其中在这些实验模型中,我们研究了可能促进癌症产生的单个因素。”

在2015年1月,来自美国约翰霍普金斯大学的一个研究团队(Science, doi:10.1126/science.1260825)在Science期刊上发表证据证实干细胞突变是癌症的主要触发因子,并且发现一种组织中的干细胞在它的一生当中经历的分裂次数能够解释不同组织类型之间大约65%的癌症风险差异。随后,在2016年1月,来自美国纽约石溪大学的Yusuf Hannun和他的同事们发现几乎所有的癌症病例是由于环境暴露(Nature, doi:10.1038/nature16166)。


近日,包括日本国家癌症中心在内的多家科研机构联合发表文章,宣布开发出一种叫做NCB-0846的小分子Wnt抑制剂。Wnt信号途径是癌症干细胞发育的一条关键途径,这种抑制剂的出现或为药物难治性结直肠癌病人提供新的治疗选择。

日本科学家们之前检测了TCF4和β-catenin形成的转录复合物,鉴定出TNIK是调节TCF4/β-catenin的一种重要的调控成分。TNIK在Wnt信号途径的最下游发挥调控作用,因此研究人员认为即使结直肠癌细胞中存在APC基因突变,对TNIK的药物学抑制仍然有望阻断该信号途径。

研究人员对一个针对激酶建立的化合物分子库进行筛选,经过优化之后发现了一种叫做NCB-0846的化合物,这种化合物能够有效抑制TNIK的激酶活性。X射线晶体结构分析表明NCB-0846与TNIK结合形成一种不活跃的构象,可能导致Wnt信号的抑制。除此之外研究结果还表明通过口服给药可以抑制人源异种移植结直肠癌小鼠模型的肿瘤生长,这种化合物还能抑制结直肠癌干细胞活性和分子标记物的表达。

研究人员表示:"我们非常高兴看到NCB-0846能有这样令人兴奋的临床前结果,特别是考虑到到目前为止靶向该途径一直存在巨大困难。我们希望能够尽快进行临床试验。"
