林金明:微流控芯片上基于核酸适配体的蛋白质检测方法研究

会议的第二天来自清华大学化学系的林金明教授做了精彩的演讲。演讲主要围绕着微流控芯片上给予核酸适配体的蛋白质检测方法研究。

演讲中林教授提到。微流控芯片技术可以继承或基本继承化学和生物等领域中所涉及的样品制备、反应、分离、检测等基本操作单元,以微通道形成网络,贯穿整个系统,具有便携、低能耗、易于制作、易于掌握等优点,易于满足生命科学对生物样品进行低剂量、更高效、高灵敏、快速分离分析的需求。利用微流控芯片够贱一种简单便携的装置用于生物标记物的检测,有望未来户外即时诊断和家庭医疗提供新的方法。生物标记物特别是蛋白质标志物进行快速灵敏的检测是临床医疗诊断、药物治疗的一个重要依据。由于微流控芯片的微型化,所需的样品量极少(<10μL)。所以灵敏度是微流控芯片上蛋白质分析的一个重要难点。

免疫分析的滚环放大技术(immuno-RCA)是一种基于免疫分析、高灵敏的核算放大技术。RCA可以在室温、恒温下进行DNA扩增,不需要特殊的一起。通过免疫反应,目标蛋白与功能性核酸配体相结合获得一段具有催化作用的G-四链体。然后,通过RCA获得一条重复序列的G-四链体。G四链体能与血红素自组装,在ABTS和过氧化氢作用下,产生不同的颜色,可用肉眼直接观察。更为重要的是,根据现行动力学模型进行的RCA反应能够直接进行半定量分析不需要过多的标准物。因此,RCA是一种适合在芯片上进行信号扩增的新技术,能及大地提高分析的敏感性和特异性,用于微量的生物大分子和生物标志的检测与研究,是未来生物芯片发展和普及使用不可缺少的检测手段。

林教授的研究以检测凝血酶为例,将微流控芯片技术和功能性核酸适配体的免疫分析技术相结合,发展了一种快速简单、高灵敏度、易于携带的装置用于生物标志物的分析。此方法在微流控芯片上平行的排列了多个微通道,并在通道上修饰具有特异性的核酸适配体目标蛋白质进行的高通量、特异性捕获和分析,降低了基质干扰。利用滚环扩增和G-四链体DNAzyme的作用,实现了信号的放大,提高分析的灵敏度,降低检测限。同时,可以通过颜色的变化,可以通孤傲肉眼进行初步的判断。
