Circulation:耗竭吲哚胺2,3-双加氧酶1明显改善心肌梗死后的心功能

缺血性心血管疾病,特别是急性心肌梗死(MI)是世界范围内导致死亡的主要原因之一。吲哚胺2,3-双加氧酶1(IDO)是催化L-色氨酸(Trp)代谢的一个限速步骤酶,也是许多病理状态的重要调节因子。

Melhem等对携带IDO特异性内皮细胞、平滑肌细胞、心肌细胞或髓系细胞缺陷的小鼠的心脏修复情况,并对这些小鼠急性心肌梗死后的表现进行评估。

结果显示,在小鼠心肌梗死后,通过IDO产生的犬尿氨酸(Kyn)被显著诱导表达。IDO基因完全缺失或药物完全抑制可限制心肌梗死后的心脏损伤和心功能不全。

在心肌梗死小鼠中,平滑肌细胞、炎性细胞或心肌细胞中IDO功能缺陷不会影响小鼠心功能和心室重构。与此截然不同的是,携带内皮细胞特异性IDO缺失的小鼠的心脏功能和心肌细胞的收缩能力均获得明显改善,而且心室重构减少

给IDO缺陷小鼠补充Kyn可以消除IDO缺失的保护作用。值得注意的是,Kyn通过芳香烃受体依赖性机制产生活性氧来加速心肌细胞凋亡。

综上,该研究结果表明,IDO可能可以作为急性心肌梗死期间的一个新的治疗靶点
