我国动物源细菌耐药性研究获重大发现

 

近日,华南农业大学兽医学院教授刘雅红团队,在持续的耐药性监测过程中,从动物身上分离出对碳青霉烯和粘菌素同时耐药的大肠杆菌,并在该菌株中发现两个耐药基因,进而提出了杂合质粒形成模型,为后续研究提供了范本。文章于近日在线发表于《自然-微生物学》(Nature Microbiology),孙坚副教授为第一作者、一年级博士生杨润时为共同第一作者,廖晓萍副教授和刘雅红教授为共同通讯作者。

据悉,碳青霉烯类抗菌药是人医临床治疗多重耐药革兰氏阴性菌感染最重要的抗菌药之一,一旦碳青霉烯类药物失效,粘菌素可作为有力补充,用作治疗多重耐药阴性菌感染的最后一道防线。但是,近几年来,随着这两类药物在临床上的广泛使用,细菌再一次突破了这两道防线。

在人类健康备受细菌耐药性问题威胁的背景下,国家兽医微生物耐药性风险评估实验室刘雅红团队在持续的耐药性监测过程中,从一只患病的宠物猫中分离一株对碳青霉烯和粘菌素同时耐药的大肠杆菌,并同进一步的分子生物学研究发现,这株大肠杆菌同时携带blaNDM-5和mcr-1两个耐药基因,且两个基因同时位于一个可接合转移的杂合质粒中。基于此,研究人员提出了杂合质粒形成的模型,为后续进一步研究相似的杂合质粒提供了可行的范本。
