Disease Models Mech:被Nature泼冷水一年后,姜黄素研究又取得2项新成果

长期以来,姜黄素被认为具有疾病治疗潜能。虽然2017年1月,Nature杂志曾发文 “泼冷水”,但一些科学家似乎并没有放弃。时隔一年,两篇新论文于近期先后发表——科学家们不仅鉴定出了支持姜黄素发挥作用的关键基因,还证实了其能改善记忆力和情绪。

姜黄素(curcumin)是从姜黄(turmeric)中分离出的一种化合物,约占姜黄成分的3%-5%。自上世纪90年代初,科学家们就开始关注这类物质,并认为它可能是导致姜黄产生多种健康促进作用的潜在组成,有望用于治疗癌症和阿尔茨海默症等疾病。

然而,尽管被寄予“厚望”,但关于姜黄素治疗疾病的实质性进展却并不多。Nature杂志曾经“泼冷水”的文章就认为,没有证据表明,姜黄素有任何特别的治疗效益。姜黄素在生理条件下是不稳定的,且不容易被身体吸收。这些特性表明,它可能并不是一个好的候选药物。此外,研究人员并没能发现有双盲、安慰剂对照临床试验证明姜黄素有“包治百病”(cure-all)的潜能。

基于这篇“警示性”的报道,一些科学家希望,能够阻止进一步的浪费研究,使研究者们意识到,有些化合物虽然非常热门,但却不太可能成药。但也有人认为,姜黄素还是值得进一步研究的。而先后出炉的这两项新成果,就进一步支持了姜黄素的治疗潜能。

论文一

1月29日,发表在Disease Models & Mechanisms上的一项研究中,来自伦敦大学皇家霍洛威学院的 Robin S. B. Williams教授带领的研究小组鉴定出了可能让姜黄素发挥治疗作用的方法,以及具有相同效果但能提供更好生物利用度的相关化合物。

研究中,科学家们使用了一种变形虫(amoeba,Dictyostelium discoideum)来调查姜黄素及其相关化合物的作用机制。最终,有两个基因被证明对姜黄素的作用是必要的,并且,先前已有研究发现,这些基因与阿尔茨海默症和癌症都相关。此外,他们还鉴定出了与姜黄素有关的、能够以相同方式起作用的、更可溶的化合物。

作者们认为,这些结果证明,姜黄素可能具有积极的影响。同时,该研究也将推动科学家们继续调查,是否姜黄素及其生物利用度更好的相关化合物能够作为一种治疗癌症和阿尔茨海默症的潜在疗法。

关于姜黄素在治疗阿尔茨海默症方面的潜能,1月19日发表于American Journal of Geriatric Psychiatry上的一项研究也有了新的发现。

研究中,来自加州大学洛杉矶分校的一个科学家小组试图调查姜黄素对正常人记忆力的影响,以及对阿尔兹海默症患者大脑中斑块、神经缠结等症状的潜在作用。

论文的第一作者兼通讯作者Gary Small带领的团队开展了为期18个月的双盲、对照试验,共招募了40名患有轻度记忆衰退的中老年人。参与者被随机分成两组,每天分别接受两次90mg的姜黄素或者安慰剂。

研究结果显示,相较对照组,姜黄素组的参与者在记忆力、注意力方面有显着提高。具体来说,服用姜黄素的人其记忆力在18个月内提高了28%,同时他们的情绪也得到了轻微改善。此外,大脑PET扫描结果显示,姜黄素组参与者的大脑杏仁核、下丘脑中的淀粉样蛋白和Tau蛋白(都是与阿尔兹海默症相关的蛋白质)累积明显少于对照组。

据悉,未来,Gary Small团队将继续开展阿尔兹海默症方面的研究,同时,他们计划扩大试验规模,以调查姜黄素抗抑郁的潜能。
