Sci Rep:恶性细胞可通过外泌体传递其恶性特征促进癌症的进展

本研究旨在了解来自高转移细胞系的外泌体是否会影响侵略性较差癌细胞的行为以及血管内皮细胞的特性。结果发现,转移性结肠癌细胞能够通过外泌体将其转移表型传递给同源原代癌细胞,此种形态转变与侵袭性行为更强相关。此外,来源于转移细胞系的外泌体(SW620Exos)导致内皮通透性增高的能力较来源于非转移性细胞系外泌体的能力强(SW480Exos)。基于SWATH定量蛋白质组学分析显示,细胞骨架相关蛋白中SW6

本研究旨在了解来自高转移细胞系的外泌体是否会影响侵略性较差癌细胞的行为以及血管内皮细胞的特性。

结果发现,转移性结肠癌细胞能够通过外泌体将其转移表型传递给同源原代癌细胞,此种形态转变与侵袭性行为更强相关。此外,来源于转移细胞系的外泌体(SW620Exos)导致内皮通透性增高的能力较来源于非转移性细胞系外泌体的能力强(SW480Exos)。


基于SWATH定量蛋白质组学分析显示,细胞骨架相关蛋白中SW620Exos明显富集,包括蛋白激活RhoA/ROCK通路,已知的可引起内皮细胞变形性能及内皮细胞连接不稳定。特别是,确定了凝血酶是靶细胞中SW620Exos诱导发生作用的关键分子,这其中研究人员还发现RhoA活性显著增加。

总之,该研究结果表明,在异构环境中侵袭性亚克隆细胞释放的外泌体可通过加速肿瘤的侵袭而促进其恶性特性的传播,由此可影响肿瘤细胞的可塑性以及内皮细胞的行为。
