PNAS:细胞色素P450环氧化活性调控炎症反应

单核细胞以及巨噬细胞在慢性炎症过程中起了关键的作用,而分泌一些脂类的调控分子是其发挥作用的主要手段之一。细胞色素P450(CYP)具有环氧化催化活性,能够对花生四烯酸等底物进行特异性修饰。

过去15年来,随着研究的逐步深入,我们了解到脂肪酸代谢产物参与调控了多种炎症的信号通路。但是CYP对炎症发生的调节作用目前仍不清楚。为了研究这一问题,来自英国伦敦学院大学的David Bishop-Bailey进行了相关研究,结果发表在最近一期的《PNAS》杂志上。

首先,作者利用zymosan A对小鼠进行处理,使其产生急性的腹膜炎。之后,作者收集了小鼠的腹水,并通过液相色谱-质谱的手段检测了其中脂类代谢产物的变化。结果显示,相比对照组小鼠,发生腹膜炎的小鼠腹水中出现的CYP衍生脂类代谢产物的数量与比重出现了明显的上升。

之后,作者通过环氧化抑制剂去抑制CYP的催化活性,并重复上述的腹膜炎诱导试验。结果显示,CYP活性的抑制能够引发明显的Ly6C阳性单核细胞的聚集以及趋化因子配体CCL2的表达。

为了验证上述抑制剂处理得出的实验结果,作者利用一种she缺失突变体小鼠进行试验。该基因表达的蛋白负责环氧化脂类产物的进一步代谢以及信号的抑制。与抑制剂处理效果相反,该小鼠体内环氧化脂类产物的水平相比野生型小鼠有明显的上升。小鼠腹水中Ly6C阳性的单核细胞数量有一定的下降,相反地,Ly6G阳性的单核细胞数量开始上升。这一结果也间接证明了之前试验得到的结论。

进一步,作者希望了解在环氧化活性受到抑制的情况下单核细胞的表型有无影响。结果显示,这部分细胞的炎症清除能力发生了明显的下降。

综上,作者证明了单核细胞在炎症反应发生过程中的正向调节活性受到其内部环氧化脂类的调控。
