这些微生物作为肠道菌群,不一定会导致人体发生疾病,反而可以影响多种生理功能,对于维持健康至关重要。一些特定菌株包括大肠杆菌是人类肠道的常住菌群,其表面表达糖分子(称为碳水化合物或聚糖)。这些聚糖可被人免疫系统识别,这导致成年个体中产生高水平的循环天然抗体。据推测,天然抗体可以针对微生物所表达的糖分子,也可以识别由病原体(寄生虫可引起人类疾病)表达的类似糖分子。Bahtiyar Yilmaz博士生,发现疟原虫的寄生虫即疟疾的病原体,表达了一种糖分子叫做 alpha-gal,其也表达在大肠杆菌菌株表面,它是人类肠道菌群的一部分。
在一系列在小鼠中进行的实验中,Bahtiyar Yilmaz发现驻留在肠道的这些细菌表达α-半乳糖苷酶,足以诱导产生天然抗体,可以识别疟原虫寄生虫表面上的相同糖分子。然后,他发现,这些抗体附着到疟原虫寄生虫表面上的alpha--半乳糖苷酶(由蚊子叮咬“接种”于皮肤中,alpha--半乳糖苷酶是疟疾传播的载体)。当这种情况发生时,抗α半乳糖苷酶的抗体会激活人体免疫系统的补体级联,它“打孔”并杀死恶性疟原虫。其实保护作用是这样的,蚊子叮咬的时候,高水平抗α半乳糖苷酶抗体设法阻止寄生虫从皮肤到血流中的过渡,并通过这样做来阻止疟疾传播。
成年个体只有一小部分才能成为感染疟原虫的寄生虫,最终感染疟疾。因此认为,与那些更易受疟疾感染的3-5岁儿童形成鲜明对比,成年人可能具有自然防御机制针对疟疾传播。研究小组确定,那些有循环抗-α-半乳糖苷酶抗体水平最低的个体也是那些最容易感染疟疾的人。
相比之下,那些具有循环抗α半乳糖苷酶抗体最高水平的个体较不易被感染发生疟疾。他们揭示了原因,为什么小婴儿是如此容易感染疟疾,可能是由于他们还没有产生针对α-半乳糖苷酶糖抗体分子的足够水平。
