大隅良典诺奖成果“细胞自噬”状态有了检测新技术

细胞自噬机制是指细胞在应对短暂生存压力时,可通过降解自身非必需成分来提供营养和能量,从而维持生命。细胞自噬也可能降解潜在毒性蛋白来阻止细胞损伤,或阻止细胞凋亡进程。细胞自噬机制与阿尔茨海默病等许多疾病相关。

据日本媒体报道,东京大学教授水岛升等人在新一期美国《分子细胞》杂志网络版上发表了有关细胞自噬机制的最新研究成果。水岛升曾在诺奖得主大隅良典的实验室工作,是大隅良典的重要助手和学生。

研究人员利用基因编辑手段使细胞产生萤光蛋白,发现随着细胞自噬的进展,萤光从蓝逐渐变为绿、黄、红三色,这样一来就可以实时测定细胞自噬的状态。在动物细胞实验中,研究人员测试了多种药物,结果发现有47种药物能促进细胞自噬,还有43种药物能阻碍细胞自噬。

研究人员表示,对于依赖细胞自噬发病的癌症类型,阻碍细胞自噬的药物可以成为有效的抗癌剂。而另外一些疾病则可以针对性地使用能诱导细胞自噬的药物。未来这一技术有望应用到和细胞自噬机制有关的药物研发中。
